Summary: This study reports several cases of ethylene oxide (EtO)-type hypersensitivity reactions in autologous donors undergoing hematopoietic progenitor cell collection (HPCC). The results showed that three autologous HPCC donors experienced EtO-type reactions within 15 minutes of initiating HPCC. The study also found that EtO reactions are rare and can be easily missed.

Summary: This study identified and investigated 35 small molecules that target hematopoietic stem cell (HSC) quiescence factors. The molecules BML-260 and TAME were found to significantly increase HSC content and activate cell cycle re-entry. These molecules promoted HSC expansion by modulating cell cycle kinetics. HSCs expanded with BML-260 showed high reconstitution capacity and increased immune cell content after xenotransplantation into immunocompromised mice.

Summary: This study retrospectively investigated the association between HLA mismatches, direction of disparities, and differences at particular HLA locus on clinical outcomes of HSCT. The results showed that mismatches at the HLA-B locus were associated with worse overall survival, absence of HLA-DRB5 was associated with a higher incidence of graft-versus-host disease (GvHD), and patients with HLA class II mismatches had a lower incidence of GvHD compared to patients with HLA class I mismatches. The findings highlight the need for further research to improve the outcomes of HSCT.

Summary: This study examined transplant outcomes using mobilized peripheral blood and FoxP3+Treg cells from mice and humans. The results showed significant elevations in stem and progenitor populations in mobilized peripheral blood. Transplant recipients who received mobilized peripheral blood with expanded Treg cells exhibited reduced GVHD and maintained graft-versus-leukemia (GVL) activity. Furthermore, the use of ex-vivo expanded Treg cells from a mobilized peripheral blood donor improved outcomes of xenogeneic GVHD.

Summary: Chromosomal aberrations in pre-HCT peripheral blood of AML patients were analyzed to identify predictors of survival post-transplant. CNLOH in chr17p for CR patients and chr13q for non-CR patients were associated with poor outcomes, with three-fold increased mortality risk even after adjusting for clinical factors. Pre-transplant CNLOH in chr13q or chr17p could be used as a predictor of poor outcomes in AML patients undergoing unrelated donor HCT, warranting further large prospective studies for validation and potential treatment strategies.
