Shedded neuronal ICAM-5 suppresses T-cell activation

Intercellular adhesion molecules (ICAMs) bind to leukocyte beta 2 integrins, which, among other functions, provide costimulatory signals for T-cell activation. ICAM-5 (telencephalin) is expressed in the soma-dendritic region of neurons of the mammalian brain. The receptor for ICAM-5 is the integrin LFA-1, a major leukocyte integrin expressed in lymphocytes and microglia. In conditions of brain ischemia, epilepsy, and encephalitis, the soluble form of ICAM-5 (siCAM-5) has been detected in physiologic fluids. Here, we report that sICAM-5 attenuates the T-cell receptor-mediated activation of T cells as demonstrated by the decreased expression of the activation markers CD69, CD40L, and CD25 (IL-2R). This effect is most clearly seen in CD45RO(LOW) (naive), and not in CD45RO(High) (memory) T cells, and is most effective early in priming, but not in the presence of strong costimula- tory signals. Furthermore, sICAM-5 promotes the mRNA expression of the cytokines TGF-beta 1 and IFN-gamma, but not TNF. The formation of sICAM-5 is promoted by activated T cells through the cleavage of ICAM-5 from neurons. This suggests that ICAM-5 is involved in immune privilege of the brain and acts as an anti-inflammatory agent.

Additionally, emotional neglect and Childhood Trauma Questionnaire total score were negatively correlated with serum brain-derived neurotrophic factor levels.

Regulation of Dynamic Cell Adhesion by Integrin-Integrin Crosstalk

Summary: Most cells express multiple integrins, which can respond to cellular functions and needs through their activation state and regulation of each other. The mechanisms of integrin crosstalk are not fully understood, but they likely involve intracellular signaling pathways shared with other cell surface receptors. Phosphorylation of integrins and key intracellular molecules play a pivotal role in integrin-cytoplasmic interactions and affect integrin activity and crosstalk. Integrin beta-chains are central in regulating crosstalk. Additionally, crosstalk may also occur between integrins and related receptors such as adhesion receptors, growth factor, and SARS-CoV-2 receptors.

The findings also showed reduced cortical thicknesses and weaker intra-relationships between monocytic genes and cortices in patients, compared to healthy controls. Enhanced Anxiety and Olfactory Microglial Activation in Early-Stage Familial Alzheimer's Disease Mouse Model

Summary: Compared to wildtype mice, 5 x FAD mice exhibited enhanced anxiety as early as 2 months old and showed increased pro-inflammatory cytokines in the olfactory bulb. The microglial activation and morphological changes were more prominent in the olfactory bulb of 2-month-old 5 x FAD mice. In the frontal cortex, pro-inflammatory cytokines were upregulated at a later stage (5-6 months old).

Summary: This study aimed to investigate the association between altered immune-inflammatory response system (IRS) and compensatory immune-regulatory reflex system (IRS/CIRS) dynamics with reduced white matter integrity in patients with schizophrenia. The results showed that the IRS/CIRS ratio was significantly higher in patients with schizophrenia compared to healthy controls, and the reduced white matter integrity was mainly observed in the genu of corpus callosum (GCC). The white matter integrity of GCC was positively associated with IRS/CIRS ratio and negatively associated with negative symptom scores.
