Summary: The microenvironment is crucial for multiple myeloma development and progression. Macrophages and the urokinase plasminogen activator receptor (uPAR) system play important roles in these processes. Inhibitors of uPAR have shown potential therapeutic effects in primary multiple myeloma cells, reducing interleukein-6 and DKK-1 levels in culture medium. However, further studies are needed to validate these preliminary results.

No correlations were found between suPAR levels and age, BMI, T2 biomarkers, CRP, or spirometric parameters.

Summary: In this study, new fluorescent probes were developed for the detection and enhanced visualization of urokinase-type plasminogen activator receptor (uPAR). These probes consist of a donor-pi-acceptor-based fluorophore conjugated with a uPAR-binding AE105 peptide. Upon specific binding to uPAR, a turn-on fluorescence signal is instantly triggered, restricting the rotational deactivation of the fluorophore. The probes were successfully applied for the imaging of uPAR overexpressed on cancer cell membranes and in a cell-based uPAR inhibitor assay.

Results showed a significant positive correlation between SUVmax and Gleason score. SUVmax could discriminate between low-risk Gleason score profiles and intermediate risk Gleason score profiles accurately.

Summary: This study identifies urokinase-type plasminogen activator receptor (uPAR) as a potential therapeutic target for diffuse-type gastric cancer (DGC), a subtype of gastric cancer with low HER2 positivity rate and insensitivity to chemotherapy and immune checkpoint inhibitors. The combination of anti-uPAR and anti-Programmed cell death protein 1 (PD-1) is shown to effectively inhibit tumor growth and prolong survival through multiple mechanisms. Furthermore, uPAR chimeric antigen receptor-expressing T cells demonstrate the ability to kill DGC cells and improve survival in preclinical models, especially when combined with PD-1 blockade therapy.
