Integrative analysis of proteomics and metabolomics of anaphylactoid reaction induced by Xuesaitong injection

Injection with natural compounds is an important method in the application of natural medicine, but its adverse drug reactions (ADRs) occur frequently, particularly the anaphylactoid reaction, which accounts for more than 77% of all reactions and has become a serious threat to public health. Here, the Xuesaitong injection (XSTI) was employed as an example to elucidate its anaphylactoid mechanism and look for potential biomarkers to assay the anaphylactoid reaction of herbal medicine injection by proteomics and metabolomics. These results disclosed that 13 differential proteins and 28 metabolites, which were further approved using the ELISA method and reference standards, respectively, were suggested as potential biomarkers to examine the anaphylactoid mechanism. The up-regulated expression of Gpx1, Sc5b9, C4d and down-regulated expression of F12, Kng1, C2 and C6 revealed that the XSTI-induced anaphylactoid reaction occurs via direct stimulation, complement and the kallikrein-kinin pathway. In addition, substances that induce an anaphylactoid effect include histamine, LTB4, uric acid and other drugs, which have been confirmed to be involved in arginine and proline metabolism, histidine metabolism, arachidonic acid metabolism purine metabolism and the TCA cycle. Furthermore, separation experiments have indicated that 10-kDa molecules of XSTI are the main allergenic factor inducing an anaphylactoid reaction. (C) 2015 Elsevier B.V. All rights reserved.

Profiling transcriptomic changes and signaling pathways in atopic dermatitis by integrative analyses on multiple databases

Summary: In this study, various datasets were analyzed to identify transcriptomic signatures and key regulators of atopic dermatitis (AD). The results revealed genes related to immune response and dermal integrity, overrepresented signaling pathways in lesional AD, and the roles of regulators in epidermal integrity and immune response. The study also suggested a potential role of B cell-mediated immune response in the pathogenesis of AD, in addition to the well-known T cell-mediated immune response. Furthermore, small molecular drugs were identified as potential therapeutic targets for AD based on the analysis.

The Research on Huanglian Jiedu Decoction against Atopic Dermatitis

Summary: By utilizing network pharmacology, Huanglian Jiedu Decoction was shown to act against atopic dermatitis through a multi-component, multi-target, and multi-pathway mode of action, significantly reducing the levels of IL-4, IL-6, and IL-10 in T lymphocytes of mice with atopic dermatitis.

High level nitrosamines in rat faeces with colorectal cancer determined by a sensitive GC-MS method

Summary: In this study, a gas chromatography-mass spectrometry method was successfully established to detect N-nitrosamines (NAs) in rat feces. The method revealed that the levels of NAS were higher in the feces of rats with colorectal cancer compared to normal rats. Additionally, a high nitrite diet was found to contribute to an abnormal increase in NAs in feces. This work provides an analytical method for further in vivo study of NAs and completes a pilot study on the relationship between NAs and colorectal cancer.

Study on the Molecular Basis of Huanglian Jiedu Decoction Against Atopic Dermatitis Integrating Chemistry, Biochemistry, and Metabolomics Strategies

Summary: Atopic dermatitis (AD) is a common chronic relapsing skin inflammation that severely impacts patients' quality of life. Huanglian Jiedu decoction (HLJDD) has been shown to have multiple pharmacological effects in the treatment of AD, particularly in inhibiting itching and enhancing immunity. Through a combination of chemistry, biochemistry, and metabolomics strategies, this study identified the active substance basis of HLJDD against AD, with a focus on components such as 40AEF and CPF.

The interaction of MD-2 with small molecules in huanglian jiedu decoction play a critical role in the treatment of sepsis

Summary: This study shows that the active components of Huanglian Jiedu Decoction (HJD) can treat sepsis by inactivating the LPS-TLR4/MD-2-NF-kappa B pathway. It is also found that the active component formulation (ACF) has similar efficacy and mechanism, reducing inflammatory cell infiltration and organ damage. Furthermore, ACF regulates certain metabolites, potentially alleviating physical damage caused by HJD.

Summary: This study established a high-purine diet-induced HN rat model and found that the pharmacokinetics of febuxostat showed accumulation in HN rats. Moreover, Astragaloside IV improved the pharmacokinetics of febuxostat in HN rats by regulating gut microbiota. These findings suggest that febuxostat might accumulate in HN patients, and Astragaloside IV could reverse the accumulation through gut microbiota regulation.

Phenylpropanoid amides from Solanum rostratum and their phytotoxic activities against Arabidopsis thaliana

Summary: In this study, phenylpropanoid amides were isolated from Solanum rostratum and their chemical structures were identified. These substances showed varying levels of inhibitory effects on seed germination and root elongation of Arabidopsis thaliana, especially compounds 2 and 4. Compounds 2 and 4 also displayed potent inhibitory effects on the antioxidant-related enzyme POD.

Exploring the Latent Mechanism of Huanglian Jiedu Decoction Formula for Anti- atopic Dermatitis by Systems Pharmacology

Summary: This study explored the real material and mechanisms of Huanglian jiedu decoction (HJD) for atopic dermatitis (AD) using network pharmacology and in vivo experiments. The results showed that HJD contains 60 bioactive compounds interacting with 169 related hub targets and participates in the pathogenesis of AD through the PI3K-Akt signaling pathway, MAPK signaling pathway, Ras signaling pathway, and Fc epsilon RI signaling pathway. In vivo experiments demonstrated that HJD reduces epidermal thickness, inflammatory cell infiltration, and downregulates key indicators in the PI3K/AKT and MAPK pathways.

Niloticin binds to MD-2 to promote anti-inflammatory pathway activation in macrophage cells

Summary: This study evaluated the potential therapeutic effects of Niloticin and its ability to bind to MD-2, a protein involved in inflammatory responses mediated by TLR4. The results showed that Niloticin could significantly decrease the levels of inflammatory cytokines induced by LPS and inhibit the activation of the LPS-TLR4/MD-2-NF-kappa B signaling pathway.

Summary: Isoimperatorin (QHS), a phytoconstituent from Angelica dahurica roots, exhibits potent anti-inflammatory activity by reducing inflammatory cytokines and mediators release and expression, inhibiting the LPS-TLR4/MD-2-NF-kappa B signaling pathway. It can serve as a potential TLR4 antagonist by blocking MD-2 binding, offering a therapeutic approach for LPS-induced inflammatory responses.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Summary: Biosorption using phosphorylated hazelnut shell waste (FHS) is a promising method for lithium removal and recovery. This study investigated the performance of a continuous-flow packed-bed column system using FHS for ultra-low level lithium removal. The results showed that the lihium biosorption capacity of the FHS column was unaffected by the bed height, but decreased with increasing flow rate.

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

An integrated approach for studying the metabolic profiling of herbal medicine in mice using high-resolution mass spectrometry and metabolomics data processing tools

Summary: Analysis of in vivo exposure to traditional Chinese medicine using mass spectrometry is helpful for identifying effective ingredients and developing new drugs. This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method utilizes stir bar sorptive extraction and multidimensional gas chromatography with flame ionization detector or mass spectrometer. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
