Summary: It has been discovered that cells with chromosome damage and defective recombinase activity can survive under appropriate oxidative stress levels. By reducing intracellular reactive oxygen species (ROS) levels, cells become sensitive to ROS rather than chronic chromosome damage. Additionally, expression of the Hsp33 protein enhances cell survival and maintains an appropriate oxidative stress level in cells with oxidative damage, rescuing cell growth.

Summary: Ovarian cancer, especially high-grade serous ovarian cancer (HGSOC), has the worst prognosis among all gynecological cancers. RAD51 expression is proposed as a biomarker of platinum resistance in HGSOC patients, which can help in selecting effective therapies for therapy-resistant patients.

Summary: The study found that single-agent antibodies against PD-L1 as maintenance therapy did not significantly benefit the overall population with metastatic breast cancer, but showed improved overall survival in patients with triple-negative breast cancer. Durvalumab may have better sensitivity in patients with CD274 amplification. Analysis of lymphocyte infiltration and homologous recombination deficiency did not predict sensitivity to durvalumab, warranting further research. Maintenance chemotherapy was more effective than durvalumab in patients with hormone receptor-positive and Her2-negative disease.

Summary: The study of the DNA damage response is crucial in cancer treatment, with PARP inhibitors achieving synthetic lethality by affecting DNA repair pathways but potentially leading to resistance. Monitoring DNA resection is vital for understanding DNA repair pathways and overcoming PARPi resistance.

Summary: This study found higher mutation rates in specific DNA repair genes in African American prostate cancer patients compared to those with European ancestry. RAD family genes and PMS2, BRCA1 genes were among the most frequently mutated DNA repair genes in African American patients. This could have important implications for specific targeted therapies.

Summary: This study identifies ATR inhibition as a potential therapeutic option for ccRCC, showing that the drug M4344 induces antiproliferative effects in ccRCC cells by inhibiting the DNA damage-sensing kinase ATR. The combination of M4344 with chemotherapeutic drugs or a poly(ADP-ribose) polymerase inhibitor demonstrates better efficacy in both mouse and human ccRCC cells.

Summary: This review discusses more accurate methods to detect homologous recombination deficiency (HRD) and how these can be applied for more personalized cancer treatment. Current methods to assess HRD are insufficient, leading to an underestimation of patients with HRD. Detection of HRD scar might be a more reliable biomarker for HRD.

Summary: A cancer targeted cascade nanosystem (CMnMPt) was constructed using Mn ions-doped mesoporous silica nanoparticles (Mn@MSN) and cisplatin prodrug (Pt(IV)), with cancer cell membrane cloaking. This nanosystem could induce ferroptosis-mediated immunogenic cell death (ICD) and enhance immunotherapy efficiency.

Summary: The text discussed the importance of homologous recombination and DNA repair for genome maintenance, as well as the impact of genetic variations in essential homologous recombination genes such as BRCA1 and BRCA2 on homologous recombination deficiency (HRD) and potential therapeutic strategies. It also highlighted the need for reliable HRD testing methods, as well as current treatment and research on synthetic lethality in HRD.

Summary: This study identified APE1 as a driver of genomic evolution in esophageal adeno-carcinoma and other solid tumors. APE1 suppression caused cell cycle arrest, impaired growth, and increased cell toxicity, while APE1 overexpression led to massive chromosomal instability and oncogenic transformation. Inhibitors targeting APE1 have the potential to treat EAC and other cancers by regulating genomic instability.

Summary: The R-loop, a three-strand nucleic acid structure, plays important regulatory roles in the meiosis process, particularly in transcriptional regulation and DNA repair. The study of R-loop in mice revealed its predominant distribution at promoter-related regions, its association with high transcription activity, and its crucial role in fertility and DSB repair. These findings highlight the significance of R-loop in spermatogenesis.

Summary: We report an enzymatic approach for the self-assembly of high-order DNA constructs using a DNA polymerase with exonuclease activity to produce ssDNA overhangs for specific sticky end cohesion. Our strategy allows for the successful construction of a variety of high-order DNA nanostructures, expanding the construction toolbox for complex DNA nanostructures and enhancing the assembly of duplex fragments in molecular cloning.

Summary: This study revealed a very low prevalence of homologous recombination repair (HRR) gene mutations and homologous recombination deficiency (HRD) in ovarian clear cell carcinoma (OCCC). TP53 mutations and microsatellite instability-high (MSI-H) were uncommon, while POLE mutations were extremely rare in OCCC. The evaluation of HRR gene mutations, HRD status, POLE mutations, and MSI-H may have limited clinical significance for OCCC treatment and prognostic stratification.
