Immunohistochemical localization of C1 subunit of V-ATPase (ATPase C1) in oral squamous cell cancer and normal oral mucosa

The ATP6V1C1 gene encodes the C1 subunit of the vacuolar-ATPase (V-ATPase) proton pump. This gene is over-expressed in oral squamous cell carcinoma (OSCC) as determined by real-time quantitative polymerase chain reaction. The aim of our study was to perform an immunohistochemical study of the distribution of the C1 subunit in normal epithelium of the oral cavity and in OSCC. We analyzed the expression of the C1 subunit in eight OSCC samples and two normal oral mucosa samples using polyclonal V-ATPase C1 antibody (clone H-300). In the normal oral mucosa samples, C1 subunit staining was observed in the basal and intermediate layers of the epithelium. No staining was visible in the keratinized superficial layers. More intense staining was observed in the OSCC samples, with the predominant expression at the periphery of tumor nests and absence of expression in dyskeratotic areas. C1 subunit expression in tumor cells was predominantly cytoplasmic, although there was perinuclear and nuclear expression in some samples. These findings demonstrate that V-ATPase is necessary for proper epithelial functioning and show its importance in the development of OSCC as evidenced by the over-expression of ATP6V1C1 in OSCC.

Importance of the vaporization margin during CO2 laser treatment of oral leukoplakia: A survival study

Summary: The main objective of this study was to assess the effectiveness of CO2 laser vaporization treatment for oral leukoplakia and determine the factors that may affect lesion recurrence. A retrospective study was conducted, and it was found that there was a statistically significant association between the margin and lesion recurrence. Maintaining a safety margin of at least 3mm during CO2 laser vaporization is a key factor in preventing recurrence.

Summary: This randomized clinical trial aimed to evaluate the effect of diode laser photobiomodulation (PBM) on post-surgical healing, inflammation, and implant stability. The results showed that PBM+ group had higher healing index and lower inflammation rate, indicating that diode laser PBM can effectively reduce inflammation and improve early healing.

Summary: Deficient DNA MMR activity leads to tumors with hypermutator phenotype called microsatellite instability (MSI). MSI has become an important predictive biomarker for anti-PD-1 therapies in various tumor types. In this study, we developed and validated MSIMEP, a computational tool for predicting MSI status from microarray DNA methylation tumor profiles of colorectal cancer samples. We demonstrated the high performance of MSIMEP in predicting MSI in different colorectal cancer cohorts and tested its consistency in other tumor types. The MSIMEP models showed better performance compared to a MLH1 promoter methylation-based model in colorectal cancer.

Summary: Oral squamous cell carcinoma (OSCC) has a low survival rate and novel prognostic biomarkers are needed. Our study found that low expression of the protein E-cadherin is associated with poor prognosis in OSCC patients. Assessing E-cadherin during routine histopathology diagnoses could be a promising prognostic tool.

Genetic linkage analysis of head and neck cancer in a Spanish family

Summary: This study describes the genetic variants associated with the development of head and neck cancer (HNC) and validates their molecular implications. A family of 3 generations, where 3 members had developed HNC, was observed. It was found that 6 out of 13 members carried the same mutation of CYP26B1, and lower levels of all-trans retinoic acid (atRA) were confirmed in their plasma.

Influence of abutment shape on peri-implant tissue conditions: A randomized clinical trial

Summary: The objective of this study was to analyze the influence of 3-mm high abutments with different shapes (cylindrical abutment vs. wide abutment) on marginal bone-level changes. The results showed that the cylindrical abutment group exhibited less bone remodeling and bone loss than the wide abutment group, as well as reduced bleeding on probing. Therefore, abutment shape has a significant influence on marginal bone-level changes during the first 12 months.

Summary: This study aimed to determine the prevalence of burnout syndrome and its associated factors in the integral clinic of the Dentistry Pilot School. The results showed high levels of emotional exhaustion and depersonalization, as well as low perception of personal fulfillment among the participants. Additionally, burnout syndrome was found to be significantly related to marital status, work experience, and various physical symptoms.

Summary: Oral potentially malignant disorders (OPMD) are associated with an increased risk of oral squamous cell carcinoma (OSCC), which is characterized by aggressive features and poor prognosis. Cancer cells can reprogram their metabolism through hypoxia-inducible factor (HIF) signaling, leading to upregulation of glycometabolism-related biomarkers. This study aimed to evaluate the expression of HIF targets in OPMD and OSCC samples and identify potential correlations with clinicopathological features and prognosis.

Summary: This study found that exosomal profiles can be used as a tool for early detection of relapse and long-term outcomes in oral squamous cell carcinoma (OSCC) patients undergoing conventional therapy. The study observed a significant reduction in exosome levels after surgery and higher exosome concentrations in patients experiencing relapse. Lower exosome levels before surgery were correlated with better disease-free survival.
