Summary: Exosomes are secreted nanovesicles formed in late endosomes with the help of core proteins of the Endosomal Sorting Complex Required for Transport (ESCRT). Accessory ESCRT-III components are involved in vesicle scission but have a specific role in Rab11a-exosome generation. Knockdown of these proteins inhibits exosome production and suppresses pro-tumorigenic activities.

Summary: This study reveals that DNAJC5 is localized to the cell membrane through palmitoylation and promotes the secretion of alpha-syn in HEK293T cells. Alpha-syn can also be actively translocated and sequestered in endosomal membrane compartments in a DNAJC5-dependent manner. The palmitoylation-deficient mutation in DNAJC5 can reduce the secretion of alpha-syn, but this can be reversed by the oligomerization of DNAJC5 induced by a membrane-targeting peptide fusion. The secretion of endogenous alpha-syn mediated by DNAJC5 is also observed in human neuroblastoma cell line SH-SY5Y and human-induced pluripotent stem cell-derived midbrain dopamine neurons.

Summary: Changes in endosomes and exosomes are correlated with neurodevelopmental and neurodegenerative disorders. In mice exposed to cocaine, there is a decrease in the number of endosomes and exosomes in the brain of male mice, along with an increase in alpha-synuclein content. These changes are not observed in female mice but can be seen after ovariectomy. These findings suggest a potential mechanism for the different response of females to chronic cocaine exposure.

Summary: ALG-2 interacts with CDIP1 in a Ca2+-dependent manner, promoting the association between CDIP1 and ESCRT-I to induce cell death. CDIP1 binds to VAPA and VAPB through an FFAT-like motif at its C-terminus. Differential expression levels of ALG-2, ESCRT-I subunits, VAPA and VAPB may impact sensitivity to anticancer drugs associated with CDIP1 expression.
