A hydrogen sulphide-releasing derivative of latanoprost acid (ACS 67) was synthesized and tested in vivo to evaluate its activity on reduction of intraocular pressure and tolerability. Glutathione (GSH) and cGMP content were also measured in the aqueous humour. The increased reduction of intraocular pressure, with a marked increase of GSH and cGMP and the related potential neuroprotective properties, make this compound interesting for the treatment of glaucoma. This is the first time that an application of a hydrogen sulphide-releasing molecule is reported for the treatment of ocular diseases. (C) 2009 Elsevier Ltd. All rights reserved.

Summary: Several cases of acute non-infectious cholestatic hepatitis in Italy were linked to the consumption of Curcuma longa-containing dietary supplements. Analysis of Tuscan cases revealed a strong association between hepatotoxicity and Curcuma longa formulations with high bioavailability and dosage. A systematic review of the literature further confirmed this association, with the majority of patients having positive dechallenge outcomes.

Adverse drug reactions in SARS-CoV-2 hospitalised patients: a case-series with a focus on drug-drug interactions

Summary: This study aimed to describe the clinical and pharmacological characteristics of SARS-CoV-2 hospitalized patients, focusing on ADRs, particularly those related to drug-drug interactions (DDIs). Among the 23 Caucasian patients during the study period, most were exposed to polypharmacy, and 17.4% had comorbidities. ADRs mainly involved cardiovascular, psychiatric, and gastrointestinal disorders, with a high incidence of QT prolongation. ADRs improved or resolved completely in 60.8% of cases.

Summary: An imbalanced angiogenic growth factors and poor utero-placental perfusion are strongly correlated with preeclampsia. A refined RUPP model in C57B1/6 J mice was developed to test the efficacy of MZe786 as a potential inhibitor of soluble Flt-1 for preeclampsia therapy. The study demonstrated that MZe786 treatment successfully prevented the development of preeclampsia by reducing maternal MAP and kidney nitrotyrosine staining, and improving fetal outcomes.

Design, Synthesis and In Vitro Investigation of Novel Basic Celastrol Carboxamides as Bio-Inspired Leishmanicidal Agents Endowed with Inhibitory Activity against Leishmania Hsp90

Summary: The study utilized the natural triterpene celastrol as a lead compound for designing and synthesizing a panel of eleven CE carboxamides, which were tested in vitro for their growth inhibitory activity against Leishmania parasites. Four of these compounds showed nanomolar leishmanicidal activity and low toxicity towards human and murine cell lines. The most selective CE analogue also inhibited the ATPase activity of Leishmania protein chaperone Hsp90, suggesting potential for future biochemical investigations.

Summary: Clofazimine has been optimized to enhance its potency as a Wnt inhibitor, with MU17 identified as a more potent lead compound for targeted therapy against TNBC and Wnt-dependent cancers. MU17 displayed a 7-fold improved potency against Wnt signaling in TNBC cells and showed no adverse effects, particularly no clofazimine-related skin coloration.

Life-threatening pediatric poisoning due to ingestion of Bufo bufo toad eggs: A case report

Design of MERS-CoV entry inhibitory short peptides based on helix-stabilizing strategies

Conformationally constrained potent inhibitors for enhancer of zeste homolog 2 (EZH2)
