GC-MS method for determining faecal sterols as biomarkers of human and pastoral animal presence in freshwater sediments

In order to determine sterols and stanols in freshwater sediments to reconstruct the past presence of humans and pastoral animals, we developed an analytical method based on pressurised liquid extraction (PLE), clean-up performed using solid phase extraction (SPE) and sterol determination using gas chromatography-mass spectrometry (GCMS) analysis. PLE extraction conditions were optimised using dichloromethane (DCM) and DCM/methanol mixtures. Clean-up was performed with 2 g silica SPE cartridges, and the concentrated extracts were eluted with 70 mL DCM. Extraction yield was evaluated using an in-house reference material spiked with C-13-labelled cholesterol and aged for 10 days. In comparison with pre-extraction, where the sediment is extracted and then spiked with a known analyte concentration, this approach preserves the original composition of the sediment. DCM and DCM/methanol mixtures resulted in high extraction yields ranging from 86 to 92 % with good reproducibility (relative standard deviation (RSD) 5-8 %). PLE extraction yields obtained with DCM as the extracting solvent were about 1.5 times higher than extractions using an ultrasonic bath. The solvent extraction mixture and matrix composition strongly affected the solvent extraction composition where higher overall recoveries (70-80 %) for each compound were obtained with DCM. The extraction mixture and matrix composition also affected the analyte concentrations, resulting in a method precision ranging from 1 to 18 %. Diatomaceous earth spiked with 10 to 100 ng of sterols, and environmental samples fortified with suitable amounts of sterols provided apparent recovery values ranging from 90 to 110 %. We applied the method to environmental samples both close to and upstream from sewage discharge zones, resulting in substantially higher faecal sterol (FeSt) concentrations near the sewage. In addition, we also applied the method to a 37cm freshwater sediment core in order to evaluate its applicability for obtaining vertical sterol profiles.

Summary: The Col Margherita (MRG) Observatory is a high-altitude station located in the Eastern Italian Alps. This study investigated the seasonal and diurnal variability of total gaseous mercury (TGM) measurements from March 2018 to May 2019. The results showed that TGM concentrations were influenced by local meteorology, as well as regional and long-range transport of air masses. The lowest TGM concentrations were observed in winter, while the highest concentrations were found in spring and summer.

Assessing glyphosate in water, marine particulate matter, and sediments in the Lagoon of Venice

Summary: The development of new analytical systems and improvement of existing ones to obtain high-resolution measurements of chemical markers in ice cores is a major challenge in the paleoclimatic scientific community. This study presents an innovative combination of fast liquid chromatography coupled with tandem mass spectrometry (FLC-MS/MS) for continuous determination of organic markers in ice cores. The method achieved low detection limits and high sampling resolution, allowing for the detection of specific organic markers and suggesting the presence of forest fires.

Detection of glyphosate residues in feed, saliva, urine and faeces from a cattle farm: a pilot study

Summary: This study aimed to evaluate the occurrence of glyphosate in faeces, urine, and saliva in a cattle farm, as well as quantify its presence in feed to understand its assimilation by mammals. The results showed that all cows excreted glyphosate in their faeces, with most feeds demonstrating a non-negligible presence of the herbicide. The study also highlighted the importance of considering the environmental impact of glyphosate re-entering the environment through liquid and solid sewage spreading.

Occurrence and Characterization of Small Microplastics (<100 μm), Additives, and Plasticizers in Larvae of Simuliidae

Summary: This study is the first to investigate the ingestion of microplastics, plasticizers, additives, and particles of micro-litter by larvae of blackflies in rivers. The study found significant differences in the abundances of microplastics and plasticizers ingested by the two species under examination. Additives and plasticizers can be used as proxies for the presence of plastic polymers in the environment.

Insight into the carbonaceous fraction of three cultural layers of different age from the area of Verona (NE Italy)

Summary: Cultural layers are deposits resulting from human settlement and activity on natural soil in the past. This study examines three archaeological sites in Verona, Italy, to understand the anthropogenic influence on the development of urban Dark Earth (UDE) layers. The research analyzes the geochemistry, carbonaceous materials, and charred material to characterize the UDE layers and investigate the relationship between different variables. The results show that UDE layers formation is related to various human activities, including metal tool and ceramic manufacturing.

Airborne polar pesticides in rural and mountain sites of North-Eastern Italy: An emerging air quality issue

Summary: North-Eastern Italy, particularly the Veneto Region, is a significant agricultural and viticultural center that has experienced rapid expansion in recent years. This study aims to assess atmospheric pollution resulting from pesticide spraying in rural areas and its potential impacts on the environment and human health. A highly sensitive technique, high pressure anion exchange chromatography coupled to a triple quadrupole mass spectrometer, was used to analyze aerosol samples collected from two sites. The results revealed the presence of fourteen polar pesticides, including cyanuric acid, glyphosate, and fosetyl-aluminium, with varying concentrations in the aerosols.

Sea ice fluctuations in the Baffin Bay and the Labrador Sea during glacial abrupt climate changes

Summary: The decline of sea ice in the North Atlantic and Nordic Seas during the last glacial period, known as Dansgaard-Oeschger (D-O) events, contributed to repeated abrupt atmospheric warmings recorded in Greenland ice cores. By analyzing records from the NEEM ice core and sediment cores, researchers discovered that the replacement of stable multiyear sea ice with seasonal sea ice amplified the abrupt climate warming during D-O events, highlighting the importance of feedbacks associated with seasonal sea ice expansion in driving the North Atlantic ocean-climate system.

Wilkes subglacial basin ice sheet response to Southern Ocean warming during late Pleistocene interglacials

The results reveal extensive retreats 330,000 and 125,000 years ago, coinciding with warmer Southern Ocean temperatures and higher global mean sea level during those interglacial periods, indicating the sensitivity of the Wilkes Subglacial Basin ice sheet to ocean warming and its potential impact on sea-level change.

Chemical and visual characterisation of EGRIP glacial ice and cloudy bands within

Summary: Impurities in polar ice, especially cloudy bands with high impurity concentrations, play a critical role in ice flow, deformation, and the integrity of the ice core record. In this study, we used a combination of advanced techniques to analyze cloudy bands in glacial ice from the East Greenland Ice-core Project (EGRIP) ice core. We classified the cloudy bands into seven different types and determined the mineralogy and localization of micro-inclusions. Our results showed that the majority of minerals found are related to terrestrial dust, with quartz, feldspar, mica, and hematite being the most common. We also found carbonaceous particles, dolomite, gypsum, and rare minerals such as rutile, anatase, epidote, titanite, and grossular. The 2D impurity imaging revealed the spatial distribution of impurities in the cloudy bands, with grain boundaries containing Na, Mg, and Sr, and the grain interior containing dust-related analytes like Al, Fe, and Ti.

200-year ice core bromine reconstruction at Dome C (Antarctica): observational and modelling results

Summary: Based on a 210-year bromine record, this study finds that the bromine signal in ice at Dome C in East Antarctica is preserved and not influenced by variations in ultraviolet radiation. However, due to limited satellite measurements and low sea-ice variability in the past 30 years, the effectiveness of bromine enrichment as a proxy for past sea-ice reconstructions at Dome C is still uncertain.
