Dibenzocyclooctadiene lignans isolated from Schisandra chinensis showed antiproliferative effects in various human cancer cells. The methoxy groups at C-3, C-4, C-3', and C-4', the hydroxyl group at C-8', and the stereo-configuration of the biphenyl ring and the angeloyl group might have influence on these activities. Additional studies indicate that one of mechanism of action of an active compound schizantherin C in A549 human lung cancer cells was related to the inhibition of cell cycle progression in G0/G1 phase. Summary: This study evaluated the risk factors for incident lung cancer in patients with newly diagnosed bronchiectasis using a large nationwide database. The results showed that male sex, overweight, current smoking, living in rural areas, and comorbid chronic obstructive pulmonary disease were independently associated with a higher risk of lung cancer in individuals with bronchiectasis. On the other hand, mild alcohol consumption was negatively associated with lung cancer development in these patients.

Investigating the antibacterial mechanism of Ampelopsis cantoniensis extracts against methicillin-resistant Staphylococcus aureus via in vitro and in silico analysis

Van Ngo Thai Bich, Tan Khanh Nguyen, Thao Dao Thi Thu, Linh Thuy Thi Tran, Sa Vo Duong Nguyen, Ho Le Han, Long-Hung Dinh Pham, Trung Hoang Thanh, Van Hoa Duong, Tuan Anh Trieu, Manh Hung Tran, Phu Tran Vinh Pham

Summary: This study found that the ethyl acetate fraction extracted from Ampelopsis cantoniensis exhibited significant antibacterial activity against methicillin-resistant Staphylococcus aureus. Molecular docking and molecular dynamics methods revealed that the main compound, dihydromyricetin, could bind to the bacterial protein PBP2a at an allosteric site, thereby exerting its antibacterial effect. High performance liquid chromatography analysis confirmed dihydromyricetin as the major component of the ethyl acetate fraction. Therefore, natural products derived from Ampelopsis cantoniensis may serve as a potential therapy for methicillin-resistant Staphylococcus aureus.

Trang Thi Thu Nguyen, Trinh Thi Tuyet Nguyen, Hoang Duc Nguyen, Tan Khanh Nguyen, Phu Tran Vinh Pham, Linh Thuoc Tran, Linh Thuy Thi Tran, Manh Hung Tran

Summary: In this study, molecular docking simulation was used to evaluate the binding energy of natural secondary metabolites from Ganoderma sp. on S. aureus sortase A. The results showed that ganosinensin B and ganosinoside A had the highest binding energy and stable binding capacity to the protein. In addition, the extracts of Ganoderma sp. exhibited antibacterial activity against S. aureus. These findings provide potential candidates for developing novel drugs to treat S. aureus infection.

Isolation, structural elucidation, and cytotoxic activity investigation of novel styryl-lactone derivatives from Goniothalamus elegans: in vitro and in silico studies

Tan Khanh Nguyen, Linh Thuy Thi Tran, Trung Truong Tan, Phu Tran Vinh Pham, Linh Thuy Khanh Nguyen, Hoai Thi Nguyen, Duc Viet Ho, Manh Hung Tran

Summary: Two styryl-lactone derivatives (1 and 2) were isolated from the aerial parts of Goniothalamus elegans. Compound 1 is a newly discovered natural product, and compound 2 is reported in this plant for the first time. Both compounds showed potent cytotoxicity against cancer cell lines, with compound 1 displaying a strong binding affinity for EGFR and HER-2 proteins. ADMET predictions suggested that these compounds can be absorbed in the gastrointestinal tract and penetrate the blood-brain barrier. These findings suggest that further studies should be conducted to explore their potential as active ingredients for cancer treatment.

Design and synthesis of a library of C2-substituted sulfamidoadenosines to probe bacterial permeability

Shibin Zhao, Julian Maceren, Mia Chung, Samantha Stone, Raphael Geiben, Melissa L. Boby, Bradley S. Sherborne, Derek S. Tan

Summary: Antibiotic resistance is a major threat to public health, with Gram-negative bacteria presenting unique challenges due to their low permeability and efflux pumps. Limited understanding of the chemical rules for overcoming these barriers hinders antibacterial drug discovery. Efforts to address this issue, such as screening compound libraries and using cheminformatic analysis, have led to the design of sulfamidoadenosines with diverse substituents, showing potential utility in accumulation in Escherichia coli.

Summary: This study successfully developed a short peptide mimetic as a MERS-CoV fusion inhibitor by reproducing the key recognition features of the HR2 helix. The resulting 23-mer lipopeptide showed comparable inhibitory effect to the 36-mer HR2 peptide HR2P-M2. This has important implications for developing short peptide-based antiviral agents to treat MERS-CoV infection.
