Synthesis and biological evaluation of the Zn (II)-IDB complexes appended with oligopolyamide as potent artificial nuclease

The zinc (II) complexes, which contain oligopolyamide and bis(2-benzimidazolylmethyl) amine (IDB) conjugated by flexible linker, have been successfully synthesized, characterized, and evaluated as DNA cleavage agents. The cleavage activity of these complexes on DNA was studied by electrophoresis. The results showed that the cleavage activity of zinc (II) complexes was enhanced comparing with those without oligopolyamide. Specially, at a high reaction concentration (1.2 mM), Zn (II) complex can cleave the plasmid DNA bearing some selectivity. Further, the spectroscopic data suggested that Zn (II) complexes with oligopolyamide backbone possessed A-T (adenine and thymine) rich sequences preference. (C) 2008 Elsevier Ltd. A new oxidation-responsive nano prodrug OC-B-BBR displayed stability and ideal drug release behavior. This study demonstrated that the prodrug significantly ameliorated colitis symptoms and colon damage, regulating gut microbiota expression levels and reshaping the community.

Summary: A series of xanthone derivatives were synthesized by rhodium-catalyzed cycloaddition and sequential oxidation, demonstrating wide applicability. The derivatives showed excellent yields and different substituent electron effects, highlighting the potential of the proposed approach. The successful one-pot synthesis from 1ethynyl-2-phenoxybenzene further emphasized the attractiveness of the current method for xanthone synthesis.

Rutin-Loaded Stimuli-Responsive Hydrogel for Anti-Inflammation

Summary: An active flavonoid compound, rutin, was incorporated into a guanosine phenylborate hydrogel (GBR) for the treatment of inflammatory bowel disease (IBD). The hydrogel exhibited good stability and injectability, and achieved dual-responsive release in an acidic pH and excessive ROS-containing medium. In vivo studies showed that the GBR hydrogel had higher therapeutic efficacy than free rutin and could suppress the expression of inflammatory cytokines.

Development of a New Synthetic Route of the Key Intermediate of Irbesartan

Summary: The new synthetic route allows for the preparation of intermediate 5 of irbesartan in four steps from low-cost glycine methyl ester, avoiding the use of highly toxic cyanide. The spirocycle structure in intermediate 5 is constructed via a simple alkylation reaction with dihaloalkanes. The scale-up production process successfully yielded 332 g of intermediate 5 with high purity and overall yield, demonstrating a viable alternative strategy for irbesartan production.

Separation of avibactam by gemini quaternary ammonium salt: Extraction, precipitation, and application in large-scale production

Summary: Non-β-lactam β-lactamase inhibitors have significant clinical significance in restoring the antibacterial effect of antibiotics. In this study, a method for the separation and purification of avibactam, a non-β-lactam β-lactamase inhibitor containing a sulfonic acid group, was developed using gemini quaternary ammonium salts. The precipitation process of a specific gemini quaternary ammonium salt was successfully applied to the separation and purification of a large-scale avibactam production, providing an efficient and green approach.

Design of MERS-CoV entry inhibitory short peptides based on helix-stabilizing strategies

Summary: This study successfully developed a short peptide mimetic as a MERS-CoV fusion inhibitor by reproducing the key recognition features of the HR2 helix. The resulting 23-mer lipopeptide showed comparable inhibitory effect to the 36-mer HR2 peptide HR2P-M2. This has important implications for developing short peptide-based antiviral agents to treat MERS-CoV infection.

The chirality of compounds has a significant impact on the activity of these agonists. This study found that certain synthesized compounds showed higher glucose uptake activity. These findings provide important information for the design of novel beta(2)AR agonists for T2D treatment.

Conformationally constrained potent inhibitors for enhancer of zeste homolog 2 (EZH2)
