Factors II, V, VII and Xa have materialized as a key enzymes for the intervention of blood coagulation cascade and for the development of new anti thrombotic agents. The combined density functional quantum mechanical/molecular mechanical (QM/MM) approach has been used to access inhibition of prothrombin and thrombin production. The biological activities of coumarin derivatives as clotting factor inhibitors was quantitatively analyzed in terms of physicochemical parameters utilizing the principal component analysis. Structural requirements for maximal potency were derived from the results of a quantitative structure activity relationship analysis. (C) 2008 Elsevier Ltd. All rights reserved.

Discovery of pyridoindole derivatives as potential inhibitors for phosphodiesterase 5A: in silico and in vivo studies

Summary: This study aimed to synthesize derivatives from identified plant-based pyridoindole lead scaffold and evaluate their phosphodiesterase 5A inhibitory potential. Through in silico and in vivo studies, three promising molecules were identified as potential antihypertensive agents, with molecule D12 showing the best antihypertensive activity. This research provides a platform for designing novel biomolecules for managing hypertension by inhibiting phosphodiesterase 5A enzyme.

In silico evaluation of NO donor heterocyclic vasodilators as SARS-CoV-2 Mpro protein inhibitor

Summary: This study utilized in silico screening method and the 3D structure of SARS-CoV-2 M-pro protein to discover potential inhibitor candidates and investigate their interactions with the substrate binding pocket. The findings suggest that certain furoxan derivatives can serve as both NO donor and SARS-CoV-2 M-pro protein inhibitors, offering a dual mechanism of action for the treatment of SARS-CoV-2.

Summary: Cancer has long been a major health threat, with breast cancer being the most common among women and lung cancer being the most common overall. CDK8 has been identified as a critical oncogenic driver in breast cancer, and screening found that ZINC000005854718 has high binding affinity with CDK8 and drug-like properties. Molecular dynamics simulation confirmed the stable binding of ZINC000005854718 with CDK8.

Evaluation of DHFR Inhibition and Antimicrobial Activity of Some Newly Synthesized Quinazolin-4(3H)-one Scaffold Coupled with Benzylidene and Ethylidene Amino Motifs

Summary: This article aims to synthesize a new series of 2-phenyl-3-substituted quinazolin-4(3H)-ones fused with an azomethine (-CH=N-) connection to Benzylidene and ethylidene motifs, which show potential antimicrobial and antifungal activities. The tested compounds exhibited strong inhibition of microbial DHFR and antimicrobial action, with some compounds demonstrating potent activity against both bacteria and fungi.

Remarkable anti-breast cancer activity and molecular docking studies of ferrocene tethered pyrimidobenzothiazoles and pyrimidobenzimidazoles

Summary: Two series of structurally diverse ferrocene tethered pyrimidobenzothiazoles and pyrimidobenzimidazoles were designed, synthesized, and evaluated for their in vitro anticancer activity against human breast cancer cell lines. Among the synthesized series, five molecules showed significantly higher anticancer activity (GI50 0.018-0.022 μM) against breast carcinoma cell lines MCF-7 and MDA-MB-231 compared to the standard drug doxorubicin (GI50 0.018 μM). Moreover, most of the synthesized compounds exhibited significant antioxidant activity and some demonstrated potent anti-angiogenic effects in in-ovo chick allantoic membrane assay. In-silico molecular docking analysis revealed that the target compounds inhibit heat shock protein90 (Hsp90).

Exploring anticancer potential of nintedanib conjugated magnetic nanoparticles: In-vitro and in-silico studies

Summary: In this study, magnetic nanoparticles (MNPs) conjugated with the anticancer drug Nintedanib (NIN) via acid-labile imine bond were successfully synthesized and characterized. The drug release experiment showed that NIN was released effectively in an acidic environment. Cytotoxicity tests demonstrated that NIN-conjugated MNPs exhibited high cytotoxicity against cancer cells, indicating good cancer targeting capability. Therefore, NIN-conjugated MNPs could serve as an efficient nanocarrier for cancer targeting.

Exploring biogenic chalcones as DprE1 inhibitors for antitubercular activity via in silico approach

Summary: Cases of drug-resistant tuberculosis (TB) have increased worldwide, posing a major threat to global TB control strategies and human health. The identification of new therapies is urgently needed, and this study used computational methods to screen a library of biogenic chalcones and identified ZINC000005158606 as a potential lead molecule against drug-resistant TB. Further investigations showed that ZINC000005158606 exhibited higher anti-TB activity compared to standard drugs, indicating its potential efficacy against Mycobacterium tuberculosis.

Exploration of limonoids for their broad spectrum antiviral potential via DFT, molecular docking and molecular dynamics simulation approach

Summary: Limonoids from citrus have attracted considerable research interest due to their wide range of pharmacological potential. This study focused on the computational exploration of the antiviral potential of three vital limonoids against spike proteins of SARS-CoV-2, Zika virus NS3 helicase, and dengue virus RNA dependent RNA polymerase. The results revealed that Obacunone showed satisfactory results for density functional theory, molecular docking, and MD simulation study.
