Chemopreventive Activity of Systemically Administered Curcumin on Oral Cancer in the 4-Nitroquinoline 1-Oxide Model

Curcumin has therapeutic potential in preventing several types of cancer, including colon, liver, prostate, and breast. The goal of this study was to evaluate the chemopreventive activity of systemically administered curcumin on oral carcinogenesis induced by 4-nitroquinolone-1-oxide (4-NQO). A total of 50 male albino rats, Rattus norvegicus, (Holtzman), were divided into five groups (n = 10 per group). Four of these groups were exposed to 50 ppm 4-NQO in their drinking water ad libitum for 8 or 12 weeks, two groups were treated with curcumin by oral gavage at 30 or 100 mg/kg per day, and one group was treated with corn oil (vehicle) only. The negative control group was euthanized at baseline. Tongues of all animals were removed after euthanasia and used in the subsequent analysis because the tongue is the primary site of carcinogenesis in this model. Descriptive histological analysis and immunohistochemistry for PCNA, Bcl-2, SOCS1 e-3, and STAT3 were performed to assess the oncogenic process. The gene expression of Vimentin, E-cadherin, N-cadherin, or TWIST1 was assessed using RT-qPCR as a representative of epithelial-mesenchymal transition (EMT) events. The administration of curcumin at 100 mg/kg during the 12 weeks markedly decreased the expression of PCNA, Bcl-2, SOCS1 e-3, and STAT3. Curcumin also minimized the cellular atypia under microscopic analysis and diminished the expression of the genes associated with EMT. Photoinactivation of multispecies cariogenic biofilm mediated by aluminum phthalocyanine chloride encapsulated in chitosan nanoparticles

Summary: This study aimed to characterize aluminum phthalocyanine chloride (AlClPc) encapsulated in chitosan nanoparticles (CN) and apply it in antimicrobial photodynamic therapy (aPDT) on multispecies biofilm to analyze the antimicrobial activity and lactate production after treatment. The results showed that aPDT mediated by the nanoconjugate AlClPc + CN + L was efficient against Streptococcus mutans, Lactobacillus casei, and Candida albicans biofilms, significantly reducing the viability of the microorganisms and lactate production.

Strontium ranelate improves post-extraction socket healing in rats submitted to the administration of bisphosphonates

Summary: This study evaluated the effect of strontium ranelate on post-extraction socket healing in rats treated with bisphosphonates. Galanin mediates tumor-induced immunosuppression in head and neck squamous cell carcinoma

Summary: The study found that galanin secreted by head and neck squamous cell carcinoma (HNSCC) cells exhibits immune-suppressive and pro-tumoral effects. Galanin decreases cell proliferation and expression of pro-inflammatory cytokines, while increasing apoptosis and expression of pro-tumoral cytokines/growth factors. It also suppresses T cell activation and anti-tumoral phenotypes. High expression of galanin is associated with reduced overall survival in HNSCC patients.

Testosterone replacement relieves ligature-induced periodontitis by mitigating inflammation, increasing pro-resolving markers and promoting angiogenesis in rats: A preclinical study

Summary: This study aimed to evaluate the inflammatory profile and resolution of inflammation in ligature-induced periodontal inflammation in rats with depletion and/or supraphysiological testosterone replacement. Results showed that supraphysiological testosterone treatment stimulated gingival tissue repair during ligature-induced periodontitis, and this may be related to an anti-inflammatory and pro-resolutive mechanism resulting from the modulatory effect on PGE2 and IL-10.

Influence of tumor cell-derived TGF-β on macrophage phenotype and macrophage-mediated tumor cell invasion

Summary: In oral squamous cell carcinoma, macrophages in the tumor microenvironment play a significant role in prognosis and disease survival. This study focuses on the effects of tumor cell-secreted TGF-beta on macrophage phenotype and macrophage-facilitated tumor invasion. The results show that blocking the effects of TGF-beta can affect macrophage phenotype and impact tumor cell invasion and chemotaxis differently.

Stress induced phosphoprotein 1 overexpression controls proliferation, migration and invasion and is associated with poor survival in oral squamous cell carcinoma

Summary: In this study, the expression and impact of stress induced phosphoprotein 1 (STIP1) in oral squamous cell carcinoma (OSCC) were investigated. It was found that STIP1 was highly expressed in OSCC, and its overexpression was associated with shorter survival and higher risk of recurrence. Knockdown of STIP1 reduced proliferation, migration, and invasion of tumor cells, and decreased the number of metastases in a zebrafish model. Furthermore, STIP1 was inversely expressed with miR-218-5p, and transfection of miR-218-5p mimics decreased STIP1 levels and inhibited tumor cell proliferation, migration, and invasion.

Comparison of monocytic cell lines U937 and THP-1 as macrophage models for in vitro studies

Summary: Understanding macrophage biology is important for understanding various biological processes and developing new immunomodulatory strategies. This study compared the response of macrophages derived from THP-1 and U937 cells to different polarizing conditions, and found that they have different tendencies towards M1 and M2 phenotypes, with THP-1-derived macrophages being more responsive to M1 stimuli.

In vitro antibacterial activity of green tea-loaded chitosan nanoparticles on caries-related microorganisms and dentin after Er:YAG laser caries removal

Summary: This study aimed to determine the inhibitory effects of green tea (Gt), EGCG, and nanoformulations containing chitosan (Nchi) and chitosan+green tea (Nchi+Gt) against Streptococcus mutans and Lactobacillus casei. The results showed that Gt at a 1:32 dilution inhibited the growth of S. mutans, while EGCG at a 1:4 dilution completely inhibited the growth of both S. mutans and L. casei. Nanoformulations containing Nchi completely inhibited S. mutans at a 1:4 dilution, whereas different concentrations of Nchi and Nchi+Gt were required to inhibit the growth of L. casei. The selective removal of carious lesion effectively reduced S. mutans counts, and the nanoformulations did not promote any additional antibacterial effect in the remaining dentin.

Cytotoxicity and bioactive potential of new root repair materials for use with BMP-2 transfected human osteoblast cells

Summary: Modified formulations of calcium silicate repair materials with additives have been developed to enhance handling, consistency, biocompatibility, and bioactivity. In this study, the effects of these modified materials on human osteoblastic cells were evaluated. The results showed that the modified cements did not have cytotoxic effects and promoted cell viability, gene expression, and mineralization processes.
