A better understanding of the molecular mechanisms in adipogenesis may provide new insights into adipose tissue-related diseases. Recently, microRNAs (miRNAs) have emerged as a class of epigenetic regulators of stem cell differentiation. In this study, we found that miR-540 was an essential negative regulator of adipogenic differentiation in adipose tissue-derived stromal cells (ADSCs). Lentivirus-mediated overexpression of miR-540 resulted in blockade of the expression of C/EBP-alpha and PPAR gamma, the two master transcription factors of adipogenesis, and deficient lipid accumulation, whereas inhibition of miR-540 promoted these processes. Target gen e reporter assays showed that miR-540 directly targeted the 3'-untranslated region (3'UTR) of PPAR gamma, resulting in a decrease of PPAR gamma protein expression. Collectively, these data suggest that miR-540 represents a new adipogenic inhibitor by, at least in part, targeting PPAR gamma. (C) 2015 Wiley Periodicals, Inc.

Summary: This study analyzed the epidemiological characteristics of the Trauma and Plastic Surgery Department in the West China Hospital of Stomatology. The results showed that males and children aged younger than 6 years were at the highest risk. The most common condition was maxillofacial injuries, mostly occurring on the forehead and caused by accidental falls. The proportion of maxillofacial space infections was not high, but severe cases could be life-threatening. Bleeding had diverse causes, but most cases were easily controlled.

Summary: This study isolated and characterized factors and extracellular vesicles (LF-FVs) from human lipoaspirate fluid, and evaluated their potential therapeutic efficacy. LF-FVs were enriched with proteins and extracellular vesicles, and were found to promote fibroblast proliferation and migration in vitro and enhance burn wound healing in a rat model. These findings suggest that LF-FVs could be potentially used for wound regeneration in clinical settings.

Hmmr Acts as a Key Regulator in the ADSCs Proliferation and Mitosis

Xiao Zhang, Dou Huang, Kaide Li, Chaoying Han, Hui Li, Cai Li, Lei Liu

Summary: This study analyzed the transcriptomes of 15532 ADSCs by single cell RNA-sequencing, and identified 15 cell subpopulations, including a subpopulation of CD168(+) ADSCs that played a vital role in ADSCs proliferation. The researchers also found that Hmmr was a critical gene associated with ADSCs proliferation and mitosis, and revealed that Hmmr promoted ADSCs proliferation through the extracellular signal-regulated kinase 1/2 signaling pathway. Therefore, Hmmr may be a novel target for obesity prevention.
