Summary: This study reveals the presence of an adipogenic and a structural branch of cells, known as SWAT cells, in progenitor cells from human adipose depots. The adipogenic branch is characterized by mitochondrial activity genes and is associated with fat distribution traits. The structural branch, called SWAT cells, display a multipotent phenotype and can revert back to progenitor state or differentiate into new adipogenic cells. This differentiation map provides a new perspective on adipose tissue regulation.

Summary: This study aimed to establish a novel classification method for predicting PPAR gamma ligands and modifying chemicals, and to group chemicals based on their effects on the PPAR gamma transcriptome and downstream metabolic functions. The research developed a classification system using lipid accumulation and RNA-Seq data to identify PPAR gamma agonists and sort chemicals into potential white or brite adipogens. Environmental contaminants were accurately identified as PPAR gamma ligands distinct from therapeutic activators, demonstrating the general applicability of the computational and experimental framework to classify chemicals.

Summary: Bone marrow adiposity is a complication in osteoporotic patients, and phytochemicals can alleviate it. This study evaluated the effect of libanoridin on the adipogenic differentiation of bone marrow-derived mesenchymal stromal cells. The results showed that libanoridin inhibited adipogenic differentiation through suppressing MAPK-mediated PPAR gamma signaling. Future studies will further explore the anti-adipogenic effects of libanoridin in vivo and elucidate its action mechanism.

Summary: The study found that both SCO and ROSI can promote adipogenesis and increase the expression of several peroxisome proliferator-activated receptor gamma target genes involved in lipid accumulation in the absence of MIX. However, ROSI can also enhance adipogenesis in the absence of MIX and insulin, and differentially regulates adipogenic and lipid metabolism genes compared to SCO.

Summary: This study investigates the role of miR-136 and HSD17B12 in adipogenesis of ovine SVFs and their relationship. The results suggest that miR-136 promotes proliferation and inhibits adipogenic differentiation, while HSD17B12 has the opposite effects. These findings provide insights into the regulatory mechanisms of lipid deposition in sheep.

Summary: The loss of KDM4B in mesenchymal stromal cells (MSCs) has been found to exacerbate skeletal aging and osteoporosis by reducing bone formation and increasing marrow adiposity. KDM4B plays a crucial role in coordinating beta-catenin/Smad1-mediated transcription by removing repressive H3K9me3, affecting MSC self-renewal and exhaustion. Activation of KDM4B in MSCs might be a new strategy for preventing and treating skeletal aging and osteoporosis by reversing bone-fat imbalance.

Summary: This study found that different subpopulations of adipose stem/stromal cells (ASCs) exhibit distinct physiological properties in a 3D spheroid culture. ASC spheroids from superficial subcutaneous adipose tissue (sSAT) showed the highest response to adipogenic induction, while spheroids from deep subcutaneous adipose tissue (dSAT) showed the greatest lipolytic capacity. Spheroids from superficial retinacula cutis (sRC) exhibited a greater secretory capacity.

Summary: Adipogenic differentiation of visceral adipose tissue-resident multipotent mesenchymal stromal cells is suppressed under chronic inflammatory stress by IFN gamma and TGF beta, with the central regulator being the IFN gamma/JAK2-activated STAT5 transcription factor. Adipogenesis can be rescued by disrupting the synergy between IFN gamma and TGF beta signaling pathways.
