Renalase contributes to the renal protection of delayedischaemic preconditioning via the regulation of hypoxia-inducible factor-1α

Ischaemic preconditioning (IPC) attenuates acute kidney injury (AKI) from renal ischaemia reperfusion. Renalase, an amine oxidase secreted by the proximal tubule, not only degrades circulating catecholamines but also protects against renal ischaemia reperfusion injury. Here, it has been suggested that the renoprotective effect of renal IPC is partly mediated by renalase. In a model of brief intermittent renal IPC, the increased cortex renalase expression was found to last for 48hrs. IPC significantly reduced renal tubular inflammation, necrosis and oxidative stress following renal ischaemia reperfusion injury. Such effects were attenuated by blocking renalase with an anti-renalase monoclonal antibody. We further demonstrated that renalase expression was up-regulated by hypoxia in vitro via an hypoxia-inducible factor (HIF)-1 mechanism. The IPC-induced up-regulation of renalase in vivo was also reduced by pre-treatment with an HIF-1 inhibitor, 3-(5-Hydroxymethyl-2-furyl)-1-benzyl indazole. In summary, the renoprotective effect of IPC is partly dependent on the renalase expression, which may be triggered by hypoxia via an HIF-1 mechanism. Endogenous renalase shows potential as a therapeutic agent for the prevention and treatment of AKI.

Summary: Immune and inflammatory factors are key pathophysiological mechanisms in diabetic renal injury, with noncanonical Wnt5a signaling playing a crucial role. Upregulation of Wnt5a and CD146 in DN patients is correlated with inflammation and renal impairment progression. Inhibition of Wnt5a signaling prevents inflammatory responses induced by high glucose, and CD146 plays an essential role in activating the Wnt5a pathway.

Summary: This study identified CANX, BID, NAMPT, and BIRC5 as immune-autophagy-related genes with independent prognostic value in kidney renal clear cell carcinoma. These genes were significantly correlated with immune score, immune checkpoint, iron death, methylation, and OCLR score. The risk prognostic model based on them was well constructed, and experimental results were consistent with the bioinformatics analysis.

Summary: This study found that FGF17, PRKCG, SSTR1, and SCTR were metastatic immune-related differentially expressed genes (mIR-DEGs) with independent prognostic values in renal clear cell carcinoma. These genes were significantly associated with clinicopathological factors and immune checkpoint-related genes. The risk prognostic model based on these genes showed good predictive potential.

Fibrinogen Is Associated with Prognosis of Critically Ill Patients with Sepsis: A Study Based on Cox Regression and Propensity Score Matching

Summary: This study found that fibrinogen levels are associated with the prognosis of sepsis patients. The relationship between elevated and decreased levels of fibrinogen and mortality was shown to be nonlinear. Subgroup analysis confirmed the robustness of this association. However, after adjusting for confounding factors, the association between decreased fibrinogen levels and increased inhospital mortality was denied.
