Abnormal distributions of transition metals inside the brain are potential diagnostic markers for several central nervous system diseases, including Alzheimer's disease (AD), Parkinson's disease, dementia with Lewy bodies (DLB), bipolar disorders and depression. To further explore this possibility, the total concentrations of iron, zinc, copper, manganese, aluminum, chromium and cadmium were measured in post-mortem hippocampus and amygdala tissues taken from AD, DLB and Control patients. A statistically significant near fifty percent reduction in the total copper levels of AD patients was observed in both the hippocampus and amygdala. The statistical power of the hippocampus and amygdala copper analysis was found to be 86 and 74% respectively. No statistically significant deviations in the total metal concentrations were found for zinc, manganese, chromium or aluminum. Iron was found to be increased by 38% in AD amygdala tissues, but was unchanged in AD hippocampus tissues. Accounting for differences in tissue water content, as a function of both tissue type and disease state, revealed more consistencies with previous literature. To aid in the design of future experiments, the effect sizes for all tissue types and metals studied are also presented.

A Fluorine-19 Magnetic Resonance Probe, Shiga-Y5, Downregulates Thioredoxin-Interacting Protein Expression in the Brain of a Mouse Model of Alzheimer's Disease

Summary: Thioredoxin-interacting protein (TXNIP) is involved in disease-associated functions related to oxidative stress by inhibiting the activity of thioredoxin (TXN). The fluorine-19 magnetic resonance probe Shiga-Y5 (SY5) has therapeutic effects in an Alzheimer's disease mouse model, with its derivative Shiga-Y6 (SY6) acting as a TXNIP regulator. SY5 can be a potential therapeutic compound targeting the thioredoxin system in oxidative stress-related brain diseases.

Summary: This study investigated the accumulation of FtMt and LC3 in the midbrain of patients with progressive supranuclear palsy (PSP) and proposed a hypothesis related to mitophagy processes. The findings suggested that FtMt may play a role in the progression of PSP.

Nicotinic Acetylcholine Receptors and Microglia as Therapeutic and Imaging Targets in Alzheimer's Disease

Summary: Research has found that in the early stages of Alzheimer's disease (AD), there is a decrease in choline signaling, including nicotinic acetylcholine receptors (nAChRs). Currently, there is limited understanding of AD pathophysiology, making it difficult to suppress the progression of the disease. Non-invasive diagnostic methods that can assess disease progression before the onset of symptoms are crucial, and incorporating the concept of neurotheranostics may be valuable for diagnosis and treatment.

Summary: Oligomers of the amyloid beta (A beta) protein play a critical role in the pathogenesis of Alzheimer's disease, but their structures and mechanisms are difficult to study due to their heterogeneity and instability. This study designed and synthesized trimer models with different linkers and found that some of them exhibited cytotoxicity and the ability to form protofibrillar species.

Summary: The precision of colocalization analysis is improved by 3D technology, which is more accurate than 2D. However, generating a colocalization model may result in a complex model with numerous polygons. We developed a simplified 3D colocalization model of FtMt/LC3 and evaluated different simplification algorithms and quality assessment methods. Additionally, we found that using the x-ray shader can improve the visualization of colocalization.
