Adipose tissue and the metabolic syndrome: focusing on adiponectin and several novel adipokines

The metabolic syndrome represents a cluster of metabolic risk factors that predispose an individual to an increased risk for Type 2 diabetes, cardiovascular diseases and their associated morbidity and mortality. Visceral obesity is thought to be a major culprit. Adipokines secreted from the adipose tissue are now believed to be key factors mediating the metabolic and inflammatory effects of obesity. In this review, we shall examine the evidence suggesting that several novel adipokines, adiponectin, adipocyte fatty acid-binding protein, retinol-binding protein-4 and lipocalin-2, may hold promise as important clinical biomarkers to identify individuals at risk for the metabolic syndrome and related comorbidities.

Summary: This study demonstrates the importance of adiponectin in the selection and development of T lymphocytes within thymic nurse cell complexes. Adiponectin-expressing regulatory T-cells (Treg) have the potential to improve glucose metabolism in obese mice and inhibit mammary tumor growth, suggesting them as a promising therapeutic agent against obesity-related metabolic and cancer diseases.

Summary: This study found that circulating Adipocyte fatty acid-binding protein (AFABP) levels were associated with incident heart failure (HF) hospitalization in type 2 diabetes. Additionally, the use of sodium glucose co-transporter 2 inhibitors (SGLT2i) was shown to lower the risk of HF in these patients.

Ultrasensitive detection of blood biomarkers of Alzheimer's and Parkinson's diseases: a systematic review

Summary: This study evaluated the diagnostic value of blood biomarkers for Alzheimer's (AD) and Parkinson's diseases (PD), identifying P-tau181, T-tau and Nfl as promising blood biomarkers for AD diagnosis. The raised levels of Nfl in AD cohorts and the significant increases in P-tau181 and T-tau in AD cohorts suggest the potential for using these biomarkers in noninvasive detection methods for AD and PD.

Risk assessment with gut microbiome and metabolite markers in NAFLD development

Summary: A growing body of evidence suggests that the gut microbiota plays a role in the pathogenesis of nonalcoholic fatty liver disease (NAFLD). This study found that machine learning models integrating microbial signatures were able to accurately classify individuals based on their NAFLD status and liver fat accumulation, outperforming other clinical models. These findings raise the possibility of using gut microbiota for early clinical warning of NAFLD development.

Summary: Recent evidence suggests a link between lean body mass (LBM) and cardiovascular capacity in women. This study found that leg and arm LBM were positively associated with cardiac dimensions, ventricular relaxation, and vascular resistance in women, but not in men.

Summary: We aimed to investigate the relationship of B2M, cystatin C, and LCN-2 with stroke risk in a general Chinese population. Elevated levels of serum B2M, cystatin C, and LCN-2 were significantly associated with stroke risk. These biomarkers may serve as useful tools for clinicians to assess stroke risk.

Circulating AFABP, FGF21, and PEDF Levels as Prognostic Biomarkers of Sight-threatening Diabetic Retinopathy

Summary: This study evaluated the associations of three diabetes-related biomarkers (AFABP, FGF21, and PEDF) with the development of sight-threatening diabetic retinopathy (STDR). The results showed that serum AFABP level was independently associated with incident STDR and its addition to a clinical model improved the prediction of STDR among participants without baseline retinopathy.

Summary: This study aimed to investigate the association between serum TSP2 level and kidney function decline in patients with type 2 diabetes. The results showed that serum TSP2 level was independently associated with incident eGFR decline and could predict a rapidly declining trajectory. This finding is of great importance for the renal health management of patients with diabetes.

Chronic SIRT1 supplementation in diabetic mice improves endothelial function by suppressing oxidative stress

Summary: This study aimed to investigate the therapeutic potential of chronic recombinant murine SIRT1 (rmSIRT1) supplementation to alleviate endothelial and vascular dysfunction in diabetic mice. The results showed that arterial SIRT1 levels were significantly reduced in diabetic conditions, while rmSIRT1 supplementation restored SIRT1 levels and improved endothelial function and vascular compliance. The study suggests that SIRT1 supplementation may represent a novel therapeutic strategy to prevent and treat diabetic vascular disease by enhancing eNOS activity and suppressing NOX-related oxidative stress.

Impact of healthy lifestyles on risk of hypertension in the Chinese population: finding from SHUN-CVD study
