The collaboration between the Hippo signaling pathway and the cAMP/PKA and ERK1/2 pathways ensures the precise regulation of the LH cascade, and YAP1 directly targets Areg to mediate its actions in the ovary.

Summary: Amphiregulin (AREG) is an important growth factor in female reproductive health. It is predominantly induced by luteinizing hormone (LH) and has various roles in the regulation of ovarian functions. Aberrant expression of AREG has been found to contribute to the pathogenesis of ovarian diseases. Understanding the molecular and cellular mechanisms of AREG regulation can lead to innovative approaches for fertility regulation and the prevention and treatment of ovarian diseases.

Summary: EGCG, the most abundant catechins from green tea, has been studied for its health benefits, including its role in ovarian steroidogenesis. In human granulosa-like tumor cell line KGN, EGCG upregulates StAR expression and enhances P4 production, without affecting other steroidogenesis-related enzymes. The effects of EGCG on steroidogenesis require 67LR-mediated activation of the PKA-CREB signaling pathway.

Summary: This study investigated the dynamics of mitochondria in mammalian ovaries during folliculogenesis and luteal formation. The results showed that mitochondrial networks shifted from branched structures in follicles to spherical and tubular mitochondria in large luteal cells. Additionally, mitochondrial diameter and volume increased from folliculogenesis to luteal formation, indicating mitochondrial enlargement as an event during these processes. Modulation of mitochondrial morphology and respiratory enzyme expressions may be related to tissue remodeling during luteal formation.

Summary: The study aimed to develop an optimized culture system for maintaining ovarian cell growth and functionality. By isolating granulosa cells and theca cells from female rats and adding follicle-stimulating hormone or luteinizing hormone to the basal culture media, secretion of estradiol and androstenedione hormones could be significantly enhanced. Additionally, growth differentiation factor 9, bone morphogenic protein 15, BMP7 and basic fibroblast growth factor were found to enhance granulosa cell proliferation and estradiol production.

Summary: This study investigates the expression patterns of key enzymes involved in human ovarian steroidogenesis during follicle development. The results show that steroidogenesis-related enzymes have low expression levels during early follicle development, but HSD3B2 is upregulated in granulosa cells (GCs) as follicles progress to the antral/preovulatory stage. Immunohistochemistry analysis confirms the expression of HSD3B2 in GCs and its co-localization with CYP17A1 in a subset of theca cells (TCs) surrounding small antral follicles. Additionally, the expression of CYB5A is significantly upregulated during follicle transition from the antral stage to the pre-ovulatory stage, indicating an enhanced progesterone production. The increased expression of DHCR24, LDLR, and HMGCR suggests an increased substrate availability for ovarian steroidogenesis.

Summary: In this study, it was found that AREG upregulated the expression of SPARC in KGN cells via EGFR and activated multiple signaling pathways. It was also discovered that SPARC was a downstream target gene of StAR. Additionally, the study revealed a positive correlation between SPARC protein levels and progesterone levels in follicular fluid. These findings elucidate the regulatory role of AREG on SPARC expression and uncover a novel function of SPARC in progesterone production in granulosa cells.

Summary: This study investigated the expression levels of APLN/APJ in buffalo ovarian granulose cells (GCs) and the effects of IGF1 and FSH on APLN/APJ expression. The results showed that APLN expression was highest in large-sized GCs, while APJ expression increased with follicle enlargement. IGF1 and FSH increased the expression of APLN and FSH, and IGF1 also increased the expression of APJ. Both isoforms of APLN increased the secretion of E2 and P4, potentially through upregulation of CYP19A1 and StAR expression. In conclusion, APLN may play a crucial role in steroidogenesis and follicular development in buffalo ovarian GCs.
