The mammalian Hedgehog pathway is modulated by ANP32 proteins

Anna Hupfer, Anna Brichkina, Till Adhikary, Matthias Lauth

Summary: Medulloblastoma is the most common malignant brain tumor in children, with one subgroup driven by uncontrolled Hedgehog signaling. While clinically approved compounds exclusively target the transmembrane component Smoothened, drug resistance against SMO inhibitors is encountered frequently, highlighting the importance of identifying novel Hh pathway components.

Activation of Cilia-Independent Hedgehog/GLI1 Signaling as a Novel Concept for Neuroblastoma Therapy

Anke Koeniger, Anna Brichkina, Iris Nee, Lukas Dempwolff, Anna Hupfer, Ilya Galperin, Florian Finkernagel, Andrea Nist, Thorsten Stiewe, Till Adhikary, Wibke Diederich, Matthias Lauth

Summary: Elevated GLI1 expression levels are associated with improved survival in NB patients and GLI1 overexpression exerts tumor-suppressive traits in cultured NB cells. However, NB cells are protected from increased GLI1 levels as they have lost their primary cilia and ability to transduce Hedgehog signals. An isoxazole molecule (ISX) has been identified to activate GLI1 independently of primary cilia, providing a potential therapeutic strategy for NB treatment.

DYRK3 contributes to differentiation and hypoxic control in neuroblastoma

Ekaterina Ivanova, Shrey Dharamvir Sharma, Anna Brichkina, Petra Pfefferle, Ursula Keber, Axel Pagenstecher, Matthias Lauth

Summary: In neuroblastoma, the overexpression of DYRK3 kinase is associated with decreased survival rate in patients. DYRK3 functions as a negative regulator of HIF-driven transcriptional responses in NB cells and negatively impacts cell differentiation, suggesting an oncogenic role in the etiology of NB.

Summary: Short-chain fatty acids pentanoate and butyrate can enhance the anti-tumor activity of immune cells through metabolic and epigenetic reprogramming, making them potential therapeutic agents in cancer immunotherapy.

Matrix stiffness drives stromal autophagy and promotes formation of a protumorigenic niche

Anna Hupfer, Anna Brichkina, Anke Koeniger, Corinna Keber, Carsten Denkert, Petra Pfefferle, Frederik Helmprobst, Axel Pagenstecher, Alexander Visekruna, Matthias Lauth

Summary: Increased tissue stiffness can enhance disease progression and support cancer cell growth through promoting stiffness-induced autophagy in stromal cells, mediated by Integrin alpha V signaling pathway. This reveals a potential target for disrupting tumor-stroma crosstalk and manipulating the tumor microenvironment.
