Summary: The Sonic Hedgehog (Shh) signaling pathway plays a crucial role in tumor formation, resistance to conventional therapies, tissue regeneration, and neurodegenerative disorders. Numerous non-coding RNAs (ncRNAs) and miRNAs have been identified to interact with the Shh pathway in both malignant and non-malignant conditions. This review summarizes the interactions between ncRNAs and Shh in various contexts.

Summary: The Hedgehog (Hh) signalling pathway is crucial in embryonic development and maintaining adult tissue homeostasis, and has been implicated in the pathogenesis of inflammatory bowel disease (IBD). Targeting the Hh signalling pathway may pave the way for successful IBD treatment. Understanding the complex role of Hh signalling in IBD pathogenesis could lead to the development of new effective therapies for IBD.

Summary: Basal cell carcinoma (BCC) is the most common skin malignancy, with most cases curable through local treatments but requiring specific therapies for advanced forms. Significant progress has been made in targeting the sonic hedgehog pathway and developing new therapies. While surgery remains the gold standard, treatment for recurrent and advanced tumors is evolving, and there is a need for new targeted therapies to overcome resistance.

Summary: Primary cilia are important for sensing environmental changes in mammalian cells through the Sonic Hedgehog pathway. GPR161 is a significant player in primary cilia and its unique features may affect receptor function and cAMP compartmentalisation. The recent potential pairing of GPR161 and spexin-1 requires further investigation before GPR161 is considered 'deorphanised'. The constitutive activity and unconventional regulation of GPR161 suggest that it may not require an endogenous ligand.

Summary: The study aims to provide a culturally relevant and validated instrument to assess the geriatric knowledge of healthcare providers in Vietnam. By translating and validating the instrument, the Vietnamese Knowledge about Older Patients Quiz (VKOP-Q) was developed. The pilot study indicated that healthcare providers had low scores in areas such as geriatric physiopathology, communication techniques with sensory impaired older adults, and differentiating age-related changes from abnormal changes or symptoms.

Summary: This study compared the knowledge of aging and rational geriatric pharmacotherapy among BPharm and PharmD students in Telangana state, India. The findings showed that both groups of students had inadequate knowledge in these areas, which may be attributed to a lack of geriatric content in the pharmacy curriculum and insufficient training in this field.

Summary: Elevated GLI1 expression levels are associated with improved survival in NB patients and GLI1 overexpression exerts tumor-suppressive traits in cultured NB cells. However, NB cells are protected from increased GLI1 levels as they have lost their primary cilia and ability to transduce Hedgehog signals. An isoxazole molecule (ISX) has been identified to activate GLI1 independently of primary cilia, providing a potential therapeutic strategy for NB treatment.

Summary: In neuroblastoma, the overexpression of DYRK3 kinase is associated with decreased survival rate in patients. DYRK3 functions as a negative regulator of HIF-driven transcriptional responses in NB cells and negatively impacts cell differentiation, suggesting an oncogenic role in the etiology of NB.
