Summary: Neuronal K(V)7 channels are highly sensitive to reactive oxygen species, and the S2S3 linker of the voltage sensor plays a crucial role in redox modulation of the channels. Recent structural studies have revealed interactions between this linker and the Ca2+-binding loop of calmodulin, specifically the EF3 hand, which is essential for the signaling. Disrupting Ca2+ binding to EF3 abolishes the oxidation-induced enhancement of K(V)7.4 currents. Furthermore, the loading of EF3 with Ca2+ is critical for the reorientation of the AB fork, which translates the Ca2+ signals.

Summary: Calcium serves as a crucial signaling molecule in plant intracellular signaling in response to various stimuli, with a majority of calcium-binding proteins comprising the EF-hand motif. A genome-wide analysis of the EF-hand gene family in foxtail millet identified 180 SiEF-hand genes, which were further analyzed for phylogenetic relationships, gene structure, and motifs, leading to the clustering of these genes into 13 clades with diverse domain compositions. The study also revealed tissue-specific expression patterns of some SiEF-hand genes, providing valuable insights for further characterization of calcium sensors.

Summary: EF-hand proteins containing a Ca2+-binding motif are involved in cellular regulation. In addition to Ca2+, these proteins can also interact with other metals such as Mg2+, Pb2+, and Zn2+. EFhd1 and EFhd2 are actin-binding proteins that modulate actin rearrangement through Ca2+-dependent and Ca2+-independent mechanisms. This study shows that the actin-related activities of EFhd1 and EFhd2 may also be regulated by Zn2+ through coordination within their EF-hands.

Summary: In this study, a multiple synergism model for alkynyl substrates was proposed and theoretical investigations on various multimetallic bonding modes and the corresponding synergistic activations were conducted. Nine alkynyl multisilver coordination modes were computationally screened and their reactivity was sequenced. The study also revealed a correlation between the ligand conformation adjustability and the reactivity of the alkynyl unit. This study provides a systematic evaluation of multimetallic synergism in alkyne activation and offers guidance for the future design of multimetallic catalysts.

Summary: This study assessed the CaM-binding properties of AQP4, finding two putative CaM-binding domains in the cytoplasmic regions. The N-terminal CBD of AQP4 interacted predominantly with the N-lobe of CaM, while the C-terminal AQP4 peptide interacted with both the C-and N-lobes of CaM. The data support a unique adapter protein binding mode for CaM with AQP4.

Summary: Efficient hydrogenation of terminal alkenes using molecular hydrogen as the hydrogen source is achieved with well-defined bench stable Mn(I) complexes containing an N-heterocyclic carbene-based PCP pincer ligand as the catalyst. This reaction is environmentally friendly and economically viable, utilizing an inexpensive and abundantly available non-precious metal catalyst. DFT calculations provide mechanistic insight into the catalytic reaction.

Summary: Ca2+-binding proteins are found in almost all living organisms, and different types have varying affinities for calcium binding. This study introduces two new scoring schemes to estimate and manipulate calcium binding affinities in proteins containing EF hand motifs. By altering five residues, the researchers designed a unique EF-hand loop that binds calcium with high affinity. The mutant protein showed a 600-fold increase in affinity for Ca2+ compared to the wildtype protein.
