Summary: IQSEC2 gene mutations are associated with epilepsy, autism, and intellectual disability. The primary function of IQSEC2 is to act as a guanine nucleotide exchange factor for ARF6. Mutations at amino acid residue 350 result in loss of hindrance and constitutive activation of ARF6, while a mutation at amino acid residue 359 results in constitutive hindrance and loss of the ability to activate ARF6. These studies provide a model for understanding the dysregulation of IQSEC2 Sec 7 activity by mutant IQSEC2 proteins.

Summary: This review emphasizes the importance of phospholipase in lung cancer, as its deregulation can promote tumor growth and metastasis. Despite advancements in LC treatment, the prognosis remains poor due to therapy-resistant cancer cells and distant metastasis. The identification of novel prognostic markers, such as phosphoinositides, is crucial for understanding the underlying mechanisms of LC. The review discusses the role of phosphoinositide-specific phospholipase enzymes in LC and their potential as therapeutic targets.

Summary: AMP-activated protein kinase (AMPK) functions as a noncanonical guanine-nucleotide-exchange factor (GEF) for Arf6 in a kinase-independent manner, therefore playing a crucial role in regulating cancer cell migration and invasion. The activation of Arf6 by AMPK occurs during glucose starvation and 5-aminoimidazole-4-carboxamide-1-beta-D-ribofuranoside (AICAR) treatment, involving the C-terminal regulatory domain of AMPK.

Summary: Infection of the central nervous system caused by enterovirus 71 (EV71) in hand-foot-and-mouth disease is the main cause of death. This study identified a new pathway by which EV71 enters human brain microvascular endothelial cells (HBMECs) and showed its dependence on ADP-ribosylation factor 6 (ARF6). Inhibition of ARF6 reduced EV71 infectivity in HBMECs and a specific inhibitor of ARF6 alleviated EV71 infection mortality in mice.

Summary: In this study, blocking PLD activity with the specific inhibitor FIPI or knocking down PDL1 expression by siRNA significantly impaired EGF-induced calcium release in MDA-MB-468-NEO and MDA-MB-468-HER2 cells, indicating a potential connection between PLD activity and PLC-gamma 1-mediated calcium signaling. However, the exact mechanism of how PLD activity interferes with calcium release from intracellular stores remains unclear.

Summary: The study revealed that in hepatocellular carcinoma, miR-145-5p negatively regulates cell proliferation, migration, invasion, and apoptosis by targeting ARF6, and is significantly associated with patient prognosis. These findings provide insights into key factors of HCC metastasis.

Summary: In this study, we found that the neuronal adaptor FE65 interacts with ARF6 GEF-ARNO and enhances ARNO's stimulatory effect on ARF6-mediated neurite outgrowth. Furthermore, a mutation in FE65 disrupts its interaction with ARNO, leading to a loss of its ability to regulate ARF6-mediated neurite outgrowth.

Summary: miR-28-3p is downregulated in prostate cancer cells, while ARF6 is upregulated. The overexpression of miR-28-3p inhibits cell proliferation, colony formation, migration, and invasion by targeting ARF6, suggesting it as a potential therapeutic candidate for prostate cancer.

Summary: ARF6 may play a critical role in the development of gastric cancer (GC), rendering GC cells susceptible to oxidative stress and controlling capecitabine resistance via multiple routes. However, it does not directly regulate lipid peroxidation.

Summary: Prostate cancer, a common malignant tumor in men, has a high mortality rate, especially in the metastatic stage. MiRNAs play a crucial role in PCa progression, with miR-145-5p and PLD5 being identified as important regulators of cell proliferation. The study revealed that upregulation of miR-145-5p can inhibit the oncogenic effects of PLD5 and suppress PCa cell proliferation and metastasis.
