Summary: The SARs of substitution of the central cyclohexyl ring in AH-7921 have not been well studied. In order to expand the SARs of AH-7921 analogs, AP01 was synthesized and characterized, and pharmacological tests were conducted. AP01 showed high affinity for μ-opioid receptor (MOR), κ-opioid receptor (KOR), and serotonin transporter (SERT), and exhibited antinociception in rats. However, the 4-phenyl modification in AP01 may lead to potential toxicities beyond currently approved opioid drugs.

Summary: A series of novel benzamide derivatives were designed and synthesized from the pyridazinone scaffold. Among them, (S)-17b showed potent inhibitory activity against human class I HDAC isoforms and human myelodysplastic syndrome (SKM-1) cell line in vitro. Furthermore, (S)-17b demonstrated excellent in vivo antitumor activity in SKM-1 xenograft models, and showed better efficacy on mouse models with intact immune system compared to those with thymus deficiencies. This novel compound (S)-17b may serve as a promising drug candidate for further investigation.

Summary: A series of fexaramine analogs were synthesized and evaluated, leading to the discovery of an intestine-selective FXR partial agonist. The optimized compound showed high FXR agonistic activity in the intestine and was rapidly metabolized to inactive form, reducing potential side effects. In mice, oral administration of the compound induced the expression of target genes in the intestine and showed potential therapeutic effects on liver fibrosis.

Summary: This article investigates the nucleation and growth kinetics of the organic ionic compound DAAS in different solvents to improve its crystal growth, analyzing the structural properties and vibration modes contributing to crystal growth. The linear optical properties of DAAS at molecular level, liquid and solid phases are studied using density functional theory and UV-Visible spectroscopy, while the nonlinear optical properties are predicted using density function theory and Kurtz powder test. Comparisons are made regarding the thermal stability of DAAS with other highly nonlinear optical stilbazolium derivatives.

Summary: This study evaluated the in vivo exposure of 4-HPR and developed lipid-based formulations to enhance its exposure. Pharmacokinetic analysis showed that clinical formulation achieved effective plasma concentrations, while novel formulations achieved more than 3-fold improvement. However, solubility and first-pass intestinal elimination limited the exposure, which could be improved through CYP inhibition. Simulated exposure profiles suggested that b.i.d dosage regime with clinical formulation and new formulations/CYP inhibition were viable options to increase exposure.

Summary: The experimental results show that stable growth of large and crack-free DAST crystals can be achieved using the seed crystal fixing method, with high quality crystals obtained at lower solution concentrations and reasonable growth speeds. THz waves were generated based on these crystals using the DFG method, with tunable waves in a broad range and high energy conversion efficiency. The single-pulse energy of the DAST crystal was also found to vary with crystal thickness, with crystals of 140 µm thickness having high single-pulse energy. Additionally, various properties and spectra of the crystalline samples were investigated.

Summary: Second-order nonlinear active metal-organic CdSO4 (1 and 3%)-doped DAST (CdS-DAST) single crystals were grown using the low-cost slow evaporation method. The crystals have a monoclinic structure with non-centrosymmetric space group Cc, confirmed by crystallography. The presence of Cd2+ ions in the DAST crystal was confirmed by energy-dispersive X-ray analysis. The CdS-doped DAST crystals show enhanced nonlinear activity and higher second harmonic generation (SHG) compared to pristine DAST, making them suitable for developing photonic devices.

Summary: The study reveals that Rab43 plays a role in regulating the surface expression and signaling of endogenous alpha 2-adrenergic receptors and muscarinic acetylcholine receptors in primary neurons, as well as in the transport of specific receptor subtypes to dendritic and postsynaptic locations. Additionally, Rab43's actions are cell-specific and directed by direct interaction, implying multiple delivery routes for different GPCRs in neurons.

Summary: C1orf27 plays an important role in the surface transport of G protein-coupled receptors (GPCRs), specifically in the ER-to-Golgi transport of α2A-adrenergic receptor (α2A-AR). Its depletion inhibits the export of α2A-AR and other GPCRs, providing insights into the regulation of GPCR biosynthesis and anterograde transport.

Summary: The olfactory receptor OR51E2 is ectopically expressed in prostate tissues and regulates prostate cancer progression. A study demonstrated that beta-ionone, an OR51E2 agonist, activates ERK1/2 pathway in prostate cancer cells. The activation of ERK1/2 by beta-ionone is regulated by Golgi-translocating G gamma 9 subunit, PI3K gamma, and ARF1.

Summary: By studying G protein variants that can form GPCR-G protein complexes without nucleotide release, it was found that selectivity is reduced and most GPCRs interact with most variants. These findings suggest the absence of absolute structural incompatibility between noncognate receptor-G protein pairs and support the role of transient intermediate states in coupling selectivity.

Summary: G protein G beta gamma subunits play crucial roles in G protein-coupled receptor signaling and have been targeted for disease treatments. Recent studies have discovered non-canonical activation of G beta gamma at intracellular organelles, particularly the Golgi apparatus, where it regulates cell processes and can be targeted for therapeutic interventions. Inhibiting Golgi-compartmentalized G beta gamma signaling has shown promise in attenuating cardiomyocyte hypertrophy and prostate tumorigenesis.
