Stromal COX-2 signaling activated by deoxycholic acid mediates proliferation and invasiveness of colorectal epithelial cancer cells

COX-2 is a major regulator implicated in colonic cancer. However, how COX-2 signaling affects colonic carcinogenesis at cellular level is not clear. In this article, we investigated whether activation of COX-2 signaling by deoxycholic acid (DCA) in primary human normal and cancer associated fibroblasts play a significant role in regulation of proliferation and invasiveness of colonic epithelial cancer cells. Our results demonstrated while COX-2 signaling can be activated by DCA in both normal and cancer associated fibroblasts, the level of activation of COX-2 signaling is significantly greater in cancer associated fibroblasts than that in normal fibroblasts. In addition, we discovered that the proliferative and invasive potential of colonic epithelial cancer cells were much greater when the cells were co-cultured with cancer associated fibroblasts pre-treated with DCA than with normal fibroblasts pre-treated with DCA. Moreover, COX-2 siRNA attenuated the proliferative and invasive effect of both normal and cancer associate fibroblasts pre-treated with DCA on the colonic cancer cells. Further studies indicated that the activation of COX-2 signaling by DCA is through protein kinase C signaling. We speculate that activation of COX-2 signaling especially in cancer associated fibroblasts promotes progression of colonic cancer. (C) 2012 Elsevier Inc. All rights reserved.

Impact of binocular integrated visual field defects on healthy related quality of life in glaucoma

Suppression of TGF-β1 signaling by Matrigel via FAK signaling in cultured human trabecular meshwork cells

Summary: The study showed that culturing trabecular meshwork (TM) cells under different matrix conditions resulted in varying phenotypes, with 3D Matrigel assisting in maintaining the TM phenotype and being regulated by TGF-beta. Abnormal matrices may contribute to perpetuating pathological TM phenotypes in glaucoma patients with elevated levels of TGF-beta 2.

Comparison of the SITA Faster a new visual field strategy with SITA Fast strategy

Repeated Low-Level Red-Light Therapy for Controlling Onset and Progression of Myopia-a Review

Summary: Repeated low-level red-light therapy (RLRL) has emerged as a novel scientific approach for treating myopia by enhancing cellular metabolism and promoting repair processes. RLRL therapy, regulated by cytochrome c oxidase and dopamine-enhanced release of nitric oxide, can inhibit myopia progression and improve visual impairments without adverse effects. Targeting neuronal energy metabolism may be a major strategy for neurotherapeutics of myopia, and RLRL therapy holds great potential in controlling myopia.

Imaging and anatomical parameters of the lacrimal punctum and vertical canaliculus using optical coherence tomography

Results demonstrated that OCT can accurately measure anatomical parameters in vivo and detect punctal lesions, offering an objective basis for clinical diagnosis and treatment.

Near Infrared (NIR) Light Therapy of Eye Diseases: A Review

Summary: Near infrared (NIR) light therapy, also known as photobiomodulation therapy (PBMT), has gained significant attention as a novel therapeutic approach in ophthalmology in recent years. It has been shown to increase cytochrome oxidase and superoxide dismutase activities, inducing metabolic and antioxidant effects, as well as boost cerebral blood flow and cognitive functions without adverse effects. The scientific evidence supports the use of NIR therapy for treatment of visual and neurological conditions, focusing on molecular and cellular mechanisms in eye diseases.

A nano-liposomal carrier containing p-coumaric acid for induction of targeted apoptosis on melanoma cells and kinetic modeling

The findings showed that the liposomal form of p-CA had a greater impact on the cells. Kinetic modeling indicated that the best fitting model was zero-order.

They identified two types of oscillations, sarcomeric oscillations and sarcosynced oscillations, and visualized their behavior during propagating waves. The researchers discovered the presence of sarcomeric defect holes and sarcomeric collision holes, which are important indicators for understanding the oscillation properties of sarcomeres. This finding has important implications for improving our understanding of muscle function and its regulatory mechanisms.

TP53INP2 modulates the malignant progression of colorectal cancer by reducing the inactive form of β-catenin

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

Interaction of gallium, indium, and vanadyl curcumin complexes with hen egg-white lysozyme (HEWL): Mechanistic aspects and evaluation of antiamyloidogenic activity

Summary: This study explored the inhibitory effects of gallium curcumin, indium curcumin, and vanadyl curcumin on the amyloid fibrillation of hen egg white lysozyme, as well as the binding interactions of these metal complexes with the enzyme. The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.

The effects of noise exposure on hippocampal cognition in C57BL/6 mice via transcriptomics
