Summary: In this study, it was found that EV71 induces GSDMD/NLRP3-mediated pyroptosis in SH-SY5Y cells through up-regulated miR-146a. The potential target of miR-146a was identified as CXCR4. It was observed that CXCR4 expression was regulated by miR-146a during EV71 infection, and over-expression of CXCR4 attenuated EV71-induced pyroptosis in SY-SY5Y cells. These findings reveal a previously unrecognized mechanism in which EV71 induces nervous system cell damage through regulation of miR-146a/CXCR4-mediated pyroptosis.

Summary: This study reveals the interaction between the upregulated host protein GADD34 and EV71 and provides insights into the mechanisms by which EV71 non-structural precursor protein 3CD regulates host protein expression. GADD34 is important for EV71 replication, and its key functional domains, PEST 1, 2, and 3 regions, promote EV71 replication. Additionally, GADD34 is found to promote viral IRES independently of its eIF2α phosphatase activity.

Summary: This study found that EV71 infection can enhance exosome biogenesis, and EV71 3A protein can promote exosome secretion. Further investigation revealed that VPS25, which is associated with EV71 infection, is located on exosomes and its expression correlates to exosome production. Knock-down of VPS25 decreases exosome biogenesis, thus attenuating viral replication. The exosome inhibitor GW4869 exhibits significant antiviral activity against EV71 replication.

In this study, a high-affinity neutralizing monoclonal antibody 10D8 was prepared and found to have a strong protective effect against abrin-induced toxicity.

Summary: This study found that petroleum ether extract of Tournefortia sibirica L. (PE-TS) has inhibitory effects against EV71 and CVA16 infections, with low cytotoxicity. Preliminary mechanistic studies showed that PE-TS inhibits replication of EV71 genomic RNA and synthesis of the EV71 protein. Animal experiments demonstrated that PE-TS can protect mice from lethal dose EV71 intracranial challenge. Chemical analysis identified 60 compounds in PE-TS. This study provides a potential antiviral botanical drug candidate for the development of HFMD drugs.

Summary: p-Coumaric acid (p-CA) is a plant compound with anti-cancer activities. This study designed a nano-liposomal carrier containing p-CA to enhance its effectiveness against melanoma cells. The findings showed that the liposomal form of p-CA had a greater impact on the cells. Kinetic modeling indicated that the best fitting model was zero-order.

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

Summary: This study explored the inhibitory effects of gallium curcumin, indium curcumin, and vanadyl curcumin on the amyloid fibrillation of hen egg white lysozyme, as well as the binding interactions of these metal complexes with the enzyme. The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
