Summary: To qualitatively localize HA trimers displayed on assembled tetravalent HA nanoparticles, hemagglutinantigen binding fragments (Fabs) were labeled with fluorescent (FLR) dyes as probes. The labeling profile, including labeling ratio, distribution, and site-specific labeling occupancy, was evaluated to determine the labeling efficiency. A combination of intact mass measurement and peptide mapping was used to accurately profile the labeling pattern of FLR-labeled 1H01-Fabs. The data showed site-specific and semiquantitative labeling, ensuring consistency in single-particle fluorescent imaging experiments.

Summary: In this study, esters of 2-bromo-2-methylpropanoate of poly(oxyalkylene) polymers were prepared and characterized, with a focus on the effects of solvents and cationic agents on mass spectrometry results. It was found that the analyte undergoes transformation to different end-groups when certain cationic agents are used. The well-defined bromine-terminated macromolecules obtained could serve as ATRP macroinitiators for the synthesis of diverse polymeric architectures for drug delivery bioconjugates.

Summary: We report the synthesis of a new semi-polyrotaxane (SR) architecture with a coverage ratio of about 34% by encapsulating poly(isoprene) (PI) chains into 2-hydroxypropyl-beta-cyclodextrins (HP beta CD) cavities. The complexation ability of HP beta CD with PI polymer was confirmed, and the thermal, morphological, and surface properties of the PI/HP beta CD SR were analyzed.

Reliable molecular typing methods can help to trace transmission routes and manage outbreaks.

Summary: This study used an ensemble of multiple preprocessing methods to extract critical information from complicated mass spectrometry spectral data for identifying microorganisms and predicting antibiotic resistance. The ensemble method outperformed individual methods, achieving the highest accuracy on independent testing datasets. Important peaks related to antibiotic resistance could be detected, providing valuable information for further investigation of the resistance mechanism.

Summary: In this study, the identification of 230 filamentous fungi isolates using MALDI-TOF MS in combination with the Filamentous Fungi Library 3.0 was investigated. The results showed that using the more time-consuming sample preparation methods improved species-level identification rates, and the in-house threshold for species identification was validated for routine use in the laboratory. This suggests that this commercial platform is suitable for routine diagnosis.

It was found that some bioactive compounds were transformed or leached out during the processing, while new compounds related to pigments emerged. This study provides a basis for understanding the spatial distributions and changes of compounds in garlic tissue during Laba garlic processing, which may affect the bioactivities of garlic.

Summary: p-Coumaric acid (p-CA) is a plant compound with anti-cancer activities. This study designed a nano-liposomal carrier containing p-CA to enhance its effectiveness against melanoma cells. The findings showed that the liposomal form of p-CA had a greater impact on the cells. Kinetic modeling indicated that the best fitting model was zero-order.

YRNA and tRFs expression levels were found to be decreased in metastatic compared to primary MGT cell lines.

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
