Summary: Atherosclerosis is a disease characterized by increased oxidative stress, leading to modifications of proteins and lipids in the blood vessel wall. Myeloperoxidase (MPO), an enzyme mainly found in neutrophils and monocytes, plays a crucial role in this oxidative process. Research has shown that MPO interacts with lesion derived high-density lipoprotein (HDL) and modifies its components, leading to dysfunction of HDL. Understanding the impact of MPO-induced changes on the structure and function of HDL is important for the future use of HDL in diagnostics and therapy.

Summary: The study found that psoriasis patients had impaired HDL function at baseline compared to controls, but short-term and especially intermediate-term therapy with biologics markedly reduced HDL cholesterol efflux capacity and rendered HDL pro-inflammatory. However, it increased paraoxonase and lecithin-cholesterol acyltransferase activities. All biologics caused similar changes in HDL composition, subclass distribution and cholesterol efflux capacity.

Summary: Cardiovascular disease (CVD) is the leading cause of death in Western countries. Recent epidemiology studies have shown a U-shaped relationship between high-density lipoprotein cholesterol (HDL-C) levels and CVD mortality, with increased mortality in patients with extremely high HDL-C levels. HDL-C raising therapy using nicotinic acids or CETP inhibitors has shown limited efficacy in reducing CVD events. As a result, the focus has shifted to evaluating HDL functionality as a novel biomarker for CVD risk. This review provides an overview of the current status and future perspectives on HDL functions, based on findings from basic research and clinical trials.

Summary: This study explores the association between serum PFAS concentrations and lipoprotein subclasses. After adjusting for confounders, the adjusted models found positive associations between several PFASs and cholesterol concentrations in large to medium sized HDL and medium sized LDL particles. Significant associations were observed between certain PFASs and cholesterol in large to medium LDL particle sizes among women. The study also found that most PFASs tended to be associated with a reduction in VLDL particle number and VLDL triglyceride.

Summary: Preeclampsia is associated with increased cardiovascular risk for both mother and child. This study investigated the effects of preeclampsia on lipid metabolism, HDL composition, and function in mothers and neonates. Preeclampsia was linked to dyslipidemia in mothers, including high triglycerides and low HDL-cholesterol levels. Mothers with preeclampsia showed shifts in HDL subclasses and increased levels of HDL-associated apolipoprotein C-II. Neonates born to mothers with preeclampsia had increased total cholesterol levels and reduced HDL cholesterol efflux capacity. These findings suggest that preeclampsia affects both maternal and neonatal lipoprotein metabolism.

Summary: p-Coumaric acid (p-CA) is a plant compound with anti-cancer activities. This study designed a nano-liposomal carrier containing p-CA to enhance its effectiveness against melanoma cells. The findings showed that the liposomal form of p-CA had a greater impact on the cells. Kinetic modeling indicated that the best fitting model was zero-order.

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
