Summary: This study investigated the effects of the dopamine D2/D3 receptor agonist quinpirole on the behavior, brain-derived neurotrophic factor (BDNF), and neurotransmitter levels in adult zebrafish. The results showed that quinpirole administration caused decreased locomotor activity, increased anxiety-like behaviors, memory impairment, and stereotypic swimming in zebrafish. Furthermore, quinpirole treatment also decreased the number of BDNF-immunoreactive cells and altered glutamate and serotonin levels in the zebrafish brain. These findings suggest that quinpirole administration in adult zebrafish can be a useful tool for studying the mechanisms underlying neurological disorders related to the dopaminergic system.

Summary: Recent evidence shows that the SARS-CoV-2 virus inhibits immune responses through dopamine-related mechanisms. In this study, the role of dopamine in SARS-CoV-2 replication was investigated in human lung cells, revealing that dopamine can reduce viral replication and affect the expression of D1 and D2 dopamine receptors. Additionally, dopamine can upregulate the expression of ISGs and Type-I interferons, while downregulating pro-inflammatory mediators. These findings highlight the correlation between dopamine pathways and the Type-I interferon response, which may be disrupted by SARS-CoV-2 for invasion and replication in host cells.

Summary: This study investigated the longterm effects of the dopamine D2, D3 agonist quinpirole on locomotor activities in mice using a home-cage monitoring system. The results showed that quinpirole injection increased locomotor activities during the dark phase and decreased activities during the light phase. Quinpirole also affected the circadian rest-activity rhythm of locomotor activities.

Summary: Low doses of quinpirole (0.02 mg/kg body weight) induced hypolocomotion of CHL1+/+ and CHL1-/- males and females, but led to a delayed response in CHL1-/- mice. Sulpiride (1 mg/kg body weight) affected locomotion of CHL1-/- females and social interaction of CHL1+/+ females as well as social interactions of CHL1-/- and CHL1+/+ males. Quinpirole increased novelty-seeking behavior of CHL1-/- males compared to CHL1+/+ males. Vehicle-treated CHL1-/- males and females showed enhanced working memory and reduced stress-related behavior.

Summary: The difference in the secondary binding site between D2R and D3R has been utilized to design compounds with selectivity for D3R. This study prepared a series of bitopic ligands based on Fallypride to improve the selectivity for D3R using various secondary binding fragments. The results showed that compounds with a small alkyl group containing a heteroatom exhibited improved D3R selectivity.

Summary: D3/D2 sub-specificity is a complex problem to solve due to the lack of easy structural biology of G-protein coupled receptors. This study used directed mutagenesis and basic binding experiments to explore a rational approach for the discovery of highly specific D3 ligands over D2 ligands. Stable expression of receptors in HEK cells was used to construct mutants and chimeras of D2 and D3 receptors, and their binding characteristics were analyzed using standard binding conditions. The study demonstrated the feasibility and potential of the approach.

A study found that ARN15381, a multitarget compound with FAAH inhibition and DRD3 partial agonist activity, reduced nicotine self-administration in rats, suggesting the potential clinical importance of a multitarget approach in the treatment of tobacco use disorder.
