The activation of adenosine monophosphate-activated protein kinase in rat hippocampus contributes to the rapid antidepressant effect of ketamine

Recent studies have shown a rapid, robust, and lasting antidepressant effect of ketamine that makes ketamine a promising antidepressant drug. However, the mechanisms underlying this rapid antidepressant effect remain incompletely understood. The goal of the present study was to determine whether adenosine monophosphate-activated protein kinase (AMPK) was involved in ketamine's rapid antidepressant effect during the forced swimming test (FST). In the first stage of experiment, a lower level of phosphoiylated form of AMPK (p-AMPK) in the hippocampus and a longer immobility time were observed in the depressed rats during FST; whereas ketamine reversed these changes at 30 min after the administration. In the second stage of experiment, we observed that, ketamine up-regulated the levels of p-AMPK and brain-derived neurotrophic factor (BDNF) in the hippocampus of the depressed rats. Moreover, AMPK agonist strengthened the antidepressant effect of ketamine with an up-regulation of BDNF, while AMPK antagonist attenuated the antidepressant effect of ketamine with a down-regulation of BDNF. In conclusion, our results suggest that the activation of AMPK in rat hippocampus is involved in the procedure of ketamine exerting rapid antidepressant effect through the up-regulation of BDNF. Predicting postoperative gastric cancer prognosis based on inflammatory factors and machine learning technology

Risk of esketamine anesthesia on the emergence delirium in preschool children after minor surgery: a prospective observational clinical study

Summary: This study investigated the effects of a single-dose of esketamine during anesthesia induction on emergence delirium (ED) in preschool children after minor surgery. The study found that the use of esketamine was associated with a higher incidence and score of ED, as well as a longer post-anesthesia care unit stay. However, there were no significant differences in other outcomes between the two groups. Therefore, the use of esketamine in preschool children for minor surgery should be noticed.

Dysfunction of NRG1/ErbB4 Signaling in the Hippocampus Might Mediate Long-term Memory Decline After Systemic Inflammation

Summary: Accumulating evidence suggests that many sepsis survivors experience long-term cognitive impairments after hospital discharge, causing reduced quality of life and burden for their families. This study utilized a mouse model and various techniques to investigate the role of NRG1/ErbB4 signaling, PV interneurons, and hippocampal neural oscillations in memory decline after repeated LPS injections. The results demonstrated that dysfunction of NRG1/ErbB4 signaling in the hippocampus may contribute to the long-term memory decline observed in the systemic inflammation mouse model. Targeting NRG1/ErbB4 signaling in the hippocampus could be a promising approach for preventing and treating this memory decline.

A role of GABAA receptor α1 subunit in the hippocampus for rapid-acting antidepressant-like effects of ketamine

Summary: Mouse models of depression-like behavior showed decreased levels of GABA and increased levels of glutamate in the hippocampus. Plasticity in ventral pallidal cholinergic neuron-derived circuits contributes to comorbid chronic pain-like and depression-like behaviour in male mice

Summary: The study focuses on the role of ventral pallidum (VP)-derived cholinergic circuits in hyperalgesia and depression-like behavior in a chronic pain mouse model. The cholinergic circuits are activated by pain-like stimuli and become hyperactive during persistent pain. Acute stimulation of these circuits reduces pain threshold, while inhibition of the circuits elevates pain threshold. Modulation of the VP-derived cholinergic pathway for multiple days regulates depression-like behavior in chronic pain. Therefore, these circuits are implicated in comorbid hyperalgesia and depression-like behavior in a chronic pain mouse model.

Melanin-like polydopamine nanoparticles mediating anti-inflammatory and rescuing synaptic loss for inflammatory depression therapy

Summary: Inflammatory depression is closely related to neuroinflammation. Melanin-like polydopamine nanoparticles (PDA NPs) were synthesized using nano-based drug delivery technology to overcome the low permeability of current anti-inflammatory drugs. PDA NPs, with abundant phenolic hydroxyl groups, showed excellent antioxidative properties and were effective in treating lipopolysaccharide-induced inflammatory depression in mice. PDA NPs reduced inflammation, restored synaptic loss, and exhibited good biocompatibility.

Summary: Mood disorders caused by chemotherapy have become more important as cancer patients' survival increases. This study used methotrexate to induce mood disorders in rats and found that treatment with the antioxidant N-acetylcysteine (NAC) can alleviate anxiety and depression-like behaviors, increase antioxidant capacity, reduce oxidative stress and inflammatory response, and regulate brain chemistry. The findings suggest that NAC treatment could be an effective strategy in revising the treatment for individuals suffering from chemotherapy-induced mood disorders.

Continuous high-frequency repetitive transcranial magnetic stimulation at extremely low intensity affects exploratory behavior and spatial cognition in mice

Summary: This study systematically investigated the effects of extremely low intensity HF-rTMS on cognition in mice and found that 40 Hz rTMS significantly impaired exploratory behavior and spatial memory at both 10 mT and 1 mT conditions. Additionally, 40 Hz stimulation had remarkably different effects on exploratory behavior depending on intensity, compared to 10 Hz stimulation.

Clinical effects of anodal tDCS and identifying response markers in post-traumatic stress disorder (PTSD): An open-label study

Self-esteem and cortical thickness correlate with aggression in healthy children: A surface-based analysis

Summary: Aggression can have serious consequences, but little is known about its personality and neurological origins in children. This study investigated the relationship between self-esteem, aggression, and brain structure in healthy children, and found that self-esteem was negatively associated with aggression. The study also revealed that increased cortical thickness in certain brain areas may be a potential mechanism linking low self-esteem to aggression in children.

Role of serotonin in the lack of sensitization caused by prolonged food deprivation in Aplysia

Summary: Food deprivation can lead to neurological dysfunctions, including memory impairment. This study used Aplysia as an animal model to investigate the memory deficits caused by prolonged food deprivation. The results showed that 14 days of food deprivation decreased the level of 5-HT in the hemolymph, which contributed to the lack of sensitization and its cellular correlates. However, exogenous application of 5-HT partially induced sensitization in the food deprived animals.

The effects of a dual orexin receptor antagonist on fear extinction memory and sleep in mice: Implications for exposure therapy

Summary: The study found that intervention with the dual orexin receptor antagonist suvorexant did not have the expected effects on extinction memory and sleep. Higher percentages of REM sleep were associated with poorer extinction memory recall and stronger fear responses. Additionally, the fear extinction training protocol used in this study did not lead to complete fear extinction.

A reliable Coordinated Function Index (CFI) was proposed based on these parameters and simplified for improved assessment efficacy.

Transcranial alternating current stimulation does not affect microscale learning
