Betulin Elicits Anti-Cancer Effects in Tumour Primary Cultures and Cell Lines In Vitro

Betulin is a pentacyclic triterpene found in many plant species, among others, in white birch bark. The aim of the study was in vitro characterization of the anticancer activity of betulin in a range of human tumour cell lines (neuroblastoma, rhabdomyosarcoma-medulloblastoma, glioma, thyroid, breast, lung and colon carcinoma, leukaemia and multiple myeloma), and in primary tumour cultures isolated from patients (ovarian carcinoma, cervical carcinoma and glioblastoma multiforme). In this study, we demonstrated a remarkable anti-proliferative effect of betulin in all tested tumour cell cultures. Neuroblastoma (SK-N-AS) and colon carcinoma (HT-29) were the most sensitive to the anti-proliferative effect of betulin. Furthermore, betulin altered tumour cells morphology, decreased their motility and induced apoptotic cell death. These findings demonstrate the anti-cancer potential of betulin and suggest that they may be applied as an adjunctive measure in cancer treatment.

Summary: This study investigates the interaction between the cytotoxic drug PAX and sirtuin inhibitor CAM commonly used in TNBC therapy. Through detailed pharmacodynamic analysis, it is found that their combination can enhance the anti-proliferative and pro-apoptotic effects, suggesting that combination chemotherapy may be a potential approach for treating TNBC.

Summary: This study investigated the anti-cancer effects of compounds from Coptis chinensis on gastric cancer cells and found that coptisine showed the highest efficacy. The combination of coptisine with cisplatin exhibited synergistic and additive interactions in vitro, and coptisine was safe for developing zebrafish in vivo. Further studies are warranted to explore the detailed mechanisms of these bioactive compounds in inhibiting gastric cancer cell survival.

2,4-Dinitrophenol as an Uncoupler Augments the Anthracyclines Toxicity against Prostate Cancer Cells

Summary: The study aimed to determine the ability of 2,4-DNP to sensitize prostate cancer cells with different metabolic phenotypes to the action of known anthracyclines. The results showed that the simultaneous application of 2,4-DNP with doxorubicin and epirubicin exhibited a synergistic effect in LNCaP prostate cancer cells, leading to increased oxidative stress and DNA damage.

Binge-like mephedrone treatment induces memory impairment concomitant with brain kynurenic acid reduction in mice

Summary: This study aimed to investigate the effects of mephedrone on memory processes and the involvement of kynurenic acid (KYNA). The results showed that binge-like mephedrone administration resulted in memory impairments and reduced KYNA levels in the brain. In vitro studies demonstrated no effect of mephedrone on KYNA production or enzyme activity, while exposure to mephedrone in normal human brain cells showed a modest reduction in cell viability and proliferation.

Summary: The study demonstrated the synergistic effect of C1 and NTBD with AmB, showing high antifungal potential and minimal toxicity against normal human cells. In vitro experiments showed that these compounds had low cytotoxicity on human renal proximal tubule epithelial cells and exhibited strong antioxidant activity when combined with AmB. These results suggest the potential use of these compounds in the treatment of fungal infections.

Kaiso Protein Expression Correlates with Overall Survival in TNBC Patients

Summary: Triple-negative breast cancers (TNBCs) are a type of invasive carcinoma that lack specific histological characteristics. The prognosis is poor for TNBC patients, with recurrences and deaths occurring within 3-5 years after diagnosis. This study explored the correlation between the Kaiso transcription factor and breast cancer progression using clinical data and tissue samples. The results suggest that high Kaiso protein expression is associated with better overall survival and disease-free survival, as well as premenopausal age, but further studies are needed due to the heterogeneity of TNBC and context-dependent molecular diversity of Kaiso signaling.

Summary: The study found that magnoflorine has anticancer effects against gastric cancer cells by inhibiting proliferation and inducing apoptosis. Combination treatment with docetaxel produced additional pharmacological interaction. Moreover, there was no observed toxicity of magnoflorine in a zebrafish model. These findings provide a foundation for the potential therapeutic use of magnoflorine in gastric cancer.
