The phagocytosis of apo cells in patients with systemic lupus erythematosus (SLE) has been shown to be impaired. Recognition and engulfment of dying cells are mediated by molecules and receptors that are also involved in cell adhesion. Therefore, we analysed the adhesion capabilities of MoMa from patients with SLE. We employed a colorimetric assay to quantify the adhesion of MoMa to various substrates. In addition, we evaluated the morphological characteristics of the M Phi adhesion cultures by phase contrast microscopy. We have observed various morphologic characteristics depending on the adhesive substrate. The typical spindle shaped MoMa adhered to plastic and a bigger and rounder formed MoMa adhered to immunoglobulin (Ig) coated surfaces. Quantitatively, adhesion to plastic was significantly decreased in some SLE patients. Those patients showing a defective adhesion to plastic simultaneously showed an enhanced adhesion to Ig coated surfaces. These findings suggest that there is an alteration in the integrin and/or Fc gamma R mediated stimulation of MoMa from SLE patients and this alteration may be associated with the impaired clearance of apo cells seen in this disease.

Connection between Periodontitis-Induced Low-Grade Endotoxemia and Systemic Diseases: Neutrophils as Protagonists and Targets

Ljubomir Vitkov, Luis E. Munoz, Jasmin Knopf, Christine Schauer, Hannah Oberthaler, Bernd Minnich, Matthias Hannig, Martin Herrmann

Summary: Periodontitis can lead to systemic low-grade inflammation by altering the response of hematopoietic stem and progenitor cells, increasing the risk of developing systemic diseases. Inhibiting the excessive response of neutrophils in periodontitis may help reduce or prevent the occurrence of systemic diseases.

Graphene-Induced Hyperthermia (GIHT) Combined With Radiotherapy Fosters Immunogenic Cell Death

Malgorzata J. Podolska, Xiaomei Shan, Christina Janko, Rabah Boukherroub, Udo S. Gaipl, Sabine Szunerits, Benjamin Frey, Luis E. Munoz

Summary: This study demonstrates that using graphene-induced hyperthermia (GIHT) can induce melanoma cell death and enhance anti-tumor immune response by releasing multiple DAMPs. Immunization with cells and supernatants treated with GIHT and irradiation together resulted in a discreet reduction of secondary tumor volumes.

Munoz, Janina Schoen, Simon Voelkl, Susanne Merkel, Christoph Becker, Georg Schett, Robert Gruetzmann, Elisabeth Naschberger, Martin Herrmann, Michael Stuerzl

Summary: Neutrophil extracellular traps (NETs), released by neutrophils, have been found in colorectal cancer (CRC) tissues and are associated with disease progression. This study used high-resolution microscopy to detect citrullinated NETs in colon cancer tissues and found that citrullinated NETs were significantly associated with high tumour grades and lymph node metastasis. In vitro experiments also showed that purified NETs induced filopodia formation and cell motility in CRC cell lines, indicating their potential contribution to CRC metastasis.

Neutrophils Orchestrate the Periodontal Pocket

Ljubomir Vitkov, Luis E. Munoz, Janina Schoen, Jasmin Knopf, Christine Schauer, Bernd Minnich, Martin Herrmann, Matthias Hannig

Summary: The occurrence and maintenance of periodontitis are not solely attributed to dysbiosis, but are associated with dysregulated immunity and exaggerated neutrophil responses. Abnormal reactions in neutrophil activation process can lead to gingival barrier damage, bacterial penetration, and further promote the development of periodontitis.

Receptor-Mediated NETosis on Neutrophils

Tao Chen, Yanhong Li, Rui Sun, Huifang Hu, Yi Liu, Martin Herrmann, Yi Zhao, Luis E. Munoz

Summary: Neutrophil extracellular traps (NETs) play a significant role in assisting neutrophils to capture and kill microorganisms during infection, but overproduction is linked to inflammation, autoimmune disorders, and other issues. Understanding neutrophil activation signals and NET formation is crucial for studying immune regulation.

Summary: The study found that in the cardiac tissue of patients with acute myocardial infarction, there was a significant increase in neutrophil infiltration but reduced formation of NETs. Anti-DNA-IgM Favors the Detection of NET-Associated Extracellular DNA

Han Wang, Antonia Margarethe Stehr, Jeeshan Singh, Leticija Zlatar, Arndt Hartmann, Katja Evert, Elisabeth Naschberger, Saskia von Stillfried, Peter Boor, Luis E. E. Munoz, Jasmin Knopf, Michael Stuerzl, Martin Herrmann

Summary: During inflammatory responses, neutrophils utilize various defense mechanisms, such as phagocytosis, degranulation, recruitment of immune cells, secretion of anti-microbials, and release of neutrophil extracellular traps (NETs). Detection of NETs in tissue sections can be challenging due to the high fluorescence signals emitted from condensed nuclear DNA. However, using anti-DNA-IgM antibodies can effectively detect the extended DNA patches of NETs, providing a new perspective for studying neutrophil-associated immune reactions in diseases.
