Summary: In this study, a compound of curcumin nicotinate (CN) was synthesized and loaded onto biomimetic nanosystem constructed by Prussian blue nanoparticles (PB NPs) to improve its solubility. Hyaluronic acid (HA) modification was adopted to prolong circulation time and improve the accumulation of drugs in the plaque region. The designed nano-platform showed potential for prevention and targeted therapy of atherosclerosis through specific pathways.

Summary: This study reveals the relationship between Piezo1-mediated Ca2+ influx and cellular behavior using innovative biosensors and imaging analysis. The results demonstrate that Piezo1 induces extracellular Ca2+ influx, which requires the support of caveolin, cholesterol, and the cytoskeleton. Increased cytoplasmic Ca2+ levels enhance the activity of PKA, ERK, Rac1, and ROCK, promoting cancer cell survival and migration. Furthermore, Piezo1-mediated Ca2+ influx upregulates membrane ruffling, a characteristic feature of cancer cell metastasis.

Summary: A disorder of cholesterol homeostasis initiates atherosclerosis progression. Macrophage membrane-camouflaged methotrexate nanoparticles (MM@MTX NPs) showed improved therapeutic efficacy by promoting cholesterol efflux and inhibiting foam cell formation. The NPs significantly reduced plaque area, aortic plaque, and cholesterol crystal deposition in ApoE-/- mice, and exhibited good biocompatibility.

Summary: Cell behaviour is influenced by the physical forces and mechanical properties of both cells and their microenvironment. This study explores the effect of viscosity, an important component of the cellular microenvironment, on cell behaviour. The results show that cells respond in a similar manner to elevated viscosity, with increased spread area, focal adhesion formation and turnover, traction forces, and migration speed. Cells utilize membrane ruffling to sense changes in extracellular fluid viscosity and trigger adaptive responses.

Summary: The study identified an interactome of AXL and its associations with proteins regulating actin dynamics. GAS6-mediated AXL activation triggered actin remodeling and macropinocytosis, promoting the survival of cancer cells under glutamine-deprived conditions. AXL activation by GAS6 drove invasion of cancer cells through actin-driven cytoskeletal rearrangements.

Summary: A hybrid cell is constructed for the sonodynamic effect promoted cell therapy of early atherosclerosis. The study shows that the hybrid cell accumulates in atherosclerotic plaques, reducing cholesterol accumulation and inflammation, and inducing foam cell apoptosis under ultrasound activation. The cooperation of M2 macrophages and TK membranes leads to significant elimination of atherosclerotic plaques without obvious side-effects.

Summary: Plasmodium sporozoites, the mobile forms of malaria parasites, actively drive the invasion of hepatocytes with the assistance of host cell processes. The initial interactions between sporozoites and hepatocytes induce plasma membrane ruffles and filopodia extensions, which are critical for successful infection. Rho GTPase signaling plays a crucial role in regulating these processes and increasing hepatocyte susceptibility to malaria parasite infection. This study highlights the importance of host cell signaling events in plasma membrane dynamics during malaria parasite invasion of hepatocytes.

Summary: Nonalcoholic fatty liver disease (NAFLD) is a common liver disease worldwide. Currently, there are no approved therapeutic strategies for NAFLD, and hepatic biopsy is still the gold standard for diagnosis. NAFLD is a multifactorial disease with a spectrum of conditions ranging from simple steatosis to more severe stages. Studies have highlighted the need for an NAFLD risk prediction model based on genetics, biomarkers, and metabolic disorders. Mitochondrial dysfunction plays a key role in the progression of NAFLD. Alterations in the displacement loop (D-loop) region and cell-free circulating mitochondrial DNA (mt-ccf) have been proposed as potential non-invasive biomarkers for NAFLD.

The results showed that MD was more effective than VD in promoting cholesterol efflux and reducing the concentration of pro-atherogenic lipoproteins, suggesting that MD may be a better therapeutic strategy for cardiovascular prevention.
