Anti-inflammatory and anti-oxidant properties of telmisartan in cultured human umbilical vein endothelial cells

Purpose: To study whether telmisartan, an angiotensin II (AII) receptor blocker (ARB), modulates endothelial inflammation and oxidative cell damage induced by AII-independent stimuli in cultured human umbilical vein endothelial cell (HUVEC)s. Methods: Endothelial inflammation, as reflected by increased VCAM-1 and ICAM-1 expression (ELISA), was induced by TNF-alpha, an inflammatory cytokine, and cell damage (COMET and MTT assay) by hydrogen peroxide, a reactive oxygen species. Losartan, another ARB, its active metabolites (EXP-3174, EXP-3179), dexamethasone, a synthetic steroid, and pyrrolidine dithiocarbamate (PDTC), an anti-oxidant, were the controls. The contribution of PPAR-gamma agonism was assessed through synthetic PPAR-gamma agonists and antagonists and the antagonism for All-type 1 receptor-mediated stimuli by evaluating the interference against cell death induced by AII (MTT assay), a pro-apoptotic peptide that induces oxidative stress. The in vitro scavenging properties for oxyradicals were quantified by the TOSC assay. Results: Telmisartan and PDTC reduced TNF-alpha-stimulated VCAM-1 in a concentration-dependent manner while losartan, EXP-3174, EXP-3179 and dexamethasone were ineffective. All compounds did not modify ICAM-1 expression. PPAR-gamma agonists or antagonists did not interfere with the effect of telmisartan. Both ARBs antagonized AII-induced cell death but only telmisartan reduced hydrogen peroxide-induced cell damage. Telmisartan scavenged selectively hydroxyl radicals without affecting peroxyl radicals and peroxynitrite. Conclusions: Telmisartan modulates pleiotropically TNF-alpha induced VCAM-1 expression and oxidative damage in vascular endothelium, possibly by acting as a hydroxyl radical scavenger. Those anti-inflammatory and antioxidant properties may contribute to the therapeutic effect, although the applicability of these data to the clinical situations has to be verified. Summary: Polycystic ovary syndrome (PCOS) is an endocrine systemic disorder characterized by hormonal and metabolic imbalances, leading to ovulation disorders, increased androgen levels, and the formation of ovarian cysts. The molecular mechanisms and signaling pathways underlying the pathogenesis of PCOS are still not well understood. Recent research has suggested that epigenetic modifications, influenced by factors like age, environment, lifestyle, and disease state, can alter the clinical presentation of PCOS. Genetic variants in genes involved in steroid synthesis and glucose homeostasis play a crucial role in PCOS development. Epigenetic control may also affect inflammation- and cell proliferation-related genes. Lifestyle factors, such as diet and physical activity, are important in reducing oxidative stress, inflammation, and improving metabolic and hormonal parameters in PCOS management.

Mild Cognitive Impairment (MCI) could be a potential target for intervention, where antioxidants can contribute to delaying cognitive decline and crossing the threshold for clinical dementia at a later stage.

Oxidative Stress-Related Signaling Pathways Predict Oocytes' Fertilization In Vitro and Embryo Quality

Summary: The molecular/metabolic composition of follicular fluid (FF) has a significant impact on oocyte development and fertilization. The antioxidant potential of FF is correlated with the number of retrieved/mature oocytes and fertilized ones. FF from normal fertilized oocytes exhibits elevated antioxidant capability, lower levels of pro-inflammatory molecules, and higher IL-10 concentration. Samples giving rise to top-quality embryos display a unique antioxidant capability and further decrease in pro-inflammatory molecules compared to FF from poor-quality embryos. Successful embryo implantation is associated with lower pro-inflammatory molecules and high antioxidant capability. Antioxidant capability and IL-10 show good predictive ability for fertilization and embryo quality.

Summary: Heart failure is a complex clinical syndrome characterized by different etiologies and a broad spectrum of cardiac structural and functional abnormalities. Current guidelines classify heart failure based on left ventricular ejection fraction, distinguishing between reduced and preserved ejection fraction. Heart failure can progress from asymptomatic stages to clinically manifest syndrome, and the progression is associated with prognosis. However, the understanding of the steps and risk factors leading to heart failure progression is still lacking, especially in preserved ejection fraction heart failure. Precise hemodynamic and metabolic characterization may help identify different disease trajectories and risk factors, leading to specific treatment targets.

Inflammatory disease of the costotransverse joints: US evaluation in 15 symptomatic patients
