Summary: Through systematic literature search and network meta-analysis, it was found that on-demand use of Avanafil 200 mg may be the best treatment option among PDE5 inhibitors for erectile dysfunction following nerve-sparing radical prostatectomy (NSRP). Additional randomized controlled trials are needed to further validate this finding.

Summary: This review provides an updated assessment of the crucial role played by phosphodiesterase 5 (PDE5) inhibitors in the nitric oxide/cGMP pathway and their potential in treating various diseases. It discusses the medicinal chemistry and drug discovery aspects of PDE5 inhibitors, as well as the breakthrough development of allosteric inhibitors. Furthermore, the concept of multi-targeted ligands in relation to PDE5 inhibitors is critically evaluated.

Summary: Erectile dysfunction (ED) is a common condition where men are unable to achieve or maintain a satisfactory erection for sexual performance. There are various treatment options available, with phosphodiesterase 5 (PDE5) inhibitors being the first-line treatment. This review examines the safety, efficacy, and pharmacokinetic properties of new ED drugs such as udenafil, mirodenafil, youkenafil, lodenafil, and SLx-2101.

Summary: This study aimed to investigate the effects of high-dose tadalafil on type 2 diabetes. The study found that high-dose tadalafil did not decrease whole-body insulin resistance, but it improved microcirculation, liver steatosis, and metabolic control. However, this treatment was moderately tolerated and larger trials are needed to determine the optimal treatment regimen.

Summary: This paper reviewed 97 medicinal plants with PDE5 inhibitory effect, of which 77 were selected based on their traditional use as aphrodisiacs. Isoflavones and biflavones were identified as the major active constituents responsible for action, while Icariin and Icariside II were found to be the most effective inhibitors from natural sources. Traditional medicines containing extracts of Ginkgo biloba, Kaempferia parviflora, Clerodendrum colebrookianum, Eurycoma longifolia, and Vitis vinifera were also found to inhibit the PDE5 enzyme.

Summary: This study evaluated the long-term adherence to two different formulations of levothyroxine (L-T4) replacement therapy. The results showed that the adherence was generally satisfactory, with better adherence observed in patients using the liquid formulation compared to the solid formulation. Patients using tablets were more likely to forget or be careless about taking their medication, and about half of the patients preferred tablets for lifelong treatment.

Summary: This study aimed to evaluate the performance of short stems compared to traditional hip prostheses in the obese population. The results showed that both short stems and traditional implants had significant improvements in pain and clinical scores after a mean follow-up of 42.6 months. There were no significant differences between the two groups at last follow-up. Radiological evaluations also showed good positioning and osseointegration in all cases, with no significant differences in joint geometry restoration and complications.
