Sex Differences in Factors Associated With Progression of Aortic Valve Calcification in the General Population

Axel Diederichsen, Jes Sanddal Lindholt, Jacob Eifer Moller, Oke Gerke, Lars Melholt Rasmussen, Jordi S. Dahl

Summary: This study describes the natural history of aortic valve calcification (AVC) progression in the general population and identifies sex differences in factors associated with this progression rate. The magnitude of the AVC score was the most important marker of AVC progression, but dyslipidemia was associated with progression only among men and hypertension was associated with progression only among women.

Classification of Amyloidosis by Model-Assisted Mass Spectrometry-Based Proteomics

Nicolai Bjodstrup Palstrom, Aleksandra M. Rojek, Hanne E. H. Moller, Charlotte Toftmann Hansen, Rune Matthiesen, Lars Melholt Rasmussen, Niels Abildgaard, Hans Christian Beck

Summary: Amyloidosis is a rare disease caused by the misfolding and aggregation of proteins, and accurate identification of the specific proteins is crucial for treatment choice. Mass spectrometry-based proteomics has become the preferred method for identifying the amyloidogenic protein. However, manual interpretation of the data by an expert can introduce bias. To address this, a statistical model-assisted method was developed to identify amyloid-containing biopsies and classify amyloidosis subtypes. This method successfully identified novel amyloid-associated proteins and demonstrated the unbiased and reliable classification of amyloid deposits and subtype using mass spectrometry-based data.

Haptoglobin-related protein in human plasma correlates to haptoglobin concentrations and phenotypes

Maria Klojgaard Skytthe, Anna Lahn Sorensen, Dorle Hennig, Maria Boysen Sandberg, Lars Melholt Rasmussen, Peter Hojrup, Holger J. Moller, Karsten Skjodt, Soren Kragh Moestrup, Jonas Heilskov Graversen

Summary: The concentration of Hpr in plasma is correlated to the concentration of Hp and the Hp phenotypes, but its regulation mechanism is still unknown. Hpr accounts for a small portion of the Hp/Hpr pool, but can be more significant at lower Hp levels.

Artificial intelligence assisted compositional analyses of human abdominal aortic aneurysms ex vivo

Bjarne Thorsted, Lisette Bjerregaard, Pia S. Jensen, Lars M. Rasmussen, Jes S. Lindholt, Maria Bloksgaard

Summary: This study presents an analysis framework based on artificial intelligence for high throughput analysis of histological sections of excised human abdominal aortic aneurysm (AAA). The developed deep neural network (DNN) accurately segments the AAA sections and quantifies the content of different components. However, combining the analysis with immunostained serial sections requires landmark recognition. The combination of digital pathology, DNN, and landmark registration has the potential to provide mechanistic insights into AAA and support better treatment regimes.

Summary: The selective inhibition of soluble TNF has been found to reduce the expansion of abdominal aortic aneurysms in mice, indicating its potential as a treatment option.

Treatment of Vitamin K Deficiency in Hemodialysis Patients - A Pilot Study Comparing Menaquinone-7 Tablets and a Vitamin K Rich Diet

Katrine Aagaard Lentz, Julie Vahlgren, Ditte Hansen, Mario Plebani, Maria Fusaro, Lars Melholt Rasmussen, Jette Jakobsen, Jens Jorgen Sloth, Henrik Post Hansen, Jens Rikardt Andersen

Summary: A daily MK-7 supplement for 6 weeks significantly lowers dp-ucMGP and ucOC in patients on hemodialysis, while a vitamin K rich diet does not have a significant effect. Boiled broccoli is a reasonable source of phylloquinone for patients on hemodialysis, as it retains a reasonable content of phylloquinone while extracting potassium.

A targeted multiplex mass spectrometry method for quantitation of abundant matrix and cellular proteins in formalin-fixed paraffin embedded arterial tissue

Summary: This study developed a novel mass spectrometry (MS) assay for the quantification of matrix and cytosolic proteins in FFPE samples. The assay demonstrated its utility in pathophysiological studies of cardiovascular disease in diabetes through analysis of artery specimens from diabetic and non-diabetic patients.

Summary: This study analyzed the tCO(2) and HCO3-(P,st) data of patients with HCO3-(P,st) requisition at Odense University Hospital from May 11, 2021 to June 1, 2021. The results showed that tCO(2) and HCO3-(P,st) can be interchangeably used for evaluating acid-base disorders in a hospital setting.

The association between diabetes and abdominal aortic aneurysms in men: results of two Danish screening studies, a systematic review, and a meta-analysis of population-based screening studies

Summary: A paradoxical protective effect of diabetes on the development and progression of abdominal aortic aneurysms (AAA) has been investigated through a cross-sectional study, systematic review, and meta-analysis. The results suggest a potential change in the association between diabetes and AAA over time.

Cationic proteins from eosinophils bind bone morphogenetic protein receptors promoting vascular calcification and atherogenesis

Summary: In this study, it was found that blood eosinophil count and eosinophil cationic protein (ECP) concentration are risk factors for cardiovascular diseases. Eosinophils and ECP were shown to contribute to vascular calcification and atherogenesis through the BMPR-1A/1B-Smad-1/5/8-Runx2 signaling pathway.

Nicotine Administration Augments Abdominal Aortic Aneurysm Progression in Rats

Hana Hadzikadunic, Tea Bovling Sjaelland, Jes S. Lindholt, Lasse Bach Steffensen, Hans Christian Beck, Egle Kavaliunaite, Lars Melholt Rasmussen, Jane Stubbe

Summary: In this study, the researchers investigated the effect of low-dose nicotine on the progression of elastase-induced abdominal aortic aneurysms (AAAs). They found that nicotine promotes AAA progression and reduces the activity of pro-matrix metalloproteinase 2 and MMP9 in aneurysmal tissue. However, nicotine has no effect on inflammatory response and oxidative stress. Therefore, these results do not support the use of low-dose nicotine for preventing AAA progression.
