Cytotoxic and Apoptotic-inducing Effects of Purple Rice Extracts and Chemotherapeutic Drugs on Human Cancer Cell Lines

Pigmented rice is mainly black, red, and dark purple, and contains a variety of flavones, tannin, polyphenols, sterols, tocopherols, gamma-oryzanols, amino acids, and essential oils. The present study evaluated the cytotoxic effects of purple rice extracts (PREs) combined with chemotherapeutic drugs on human cancer cells and mechanisms of cell death. Methanolic (MeOH) and dichloromethane (DCM) extracts of three cultivars of purple rice in Thailand: Doisaket (DSK), Nan and Payao (PYO), were tested and compared with white rice (KK6). Cytotoxicity was determined by 3-(4, 5-dimethyl)-2, 5-diphenyltetrazolium bromide (MTT) assay in human hepatocellular carcinoma HepG2, prostate cancer LNCaP and murine normal fibroblast NIH3T3 cells. MeOH-PYO-PRE was the most cytotoxic and inhibited HepG2 cell growth more than that of LNCaP cells but was not toxic to NIH3T3 cells. When PREs were combined with paclitaxel or vinblastine, they showed additive cytotoxic effects on HepG2 and LNCaP cells, except for MeOH-PYO-PRE which showed synergistic effects on HepG2 cells when combined with vinblastine. MeOH-PYO-PRE plus vinblastine induced HepG2 cell apoptosis with loss of mitochondrial transmembrane potential (MTP) but no ROS production. MeOH-PYO-PRE-treated HepG2 cells underwent apoptosis via caspase-9 and-3 activation. The level of gamma-oryzanol was highest in DCM-PYO-PRE (44.17 mg/g) whereas anthocyanin content was high in MeOH-PYO-PRE (5.80 mg/g). In conclusion, methanolic Payao purple rice extract was mostly toxic to human HepG2 cells and synergistically enhanced the cytotoxicity of vinblastine. Human HepG2 cell apoptosis induced by MeOH-PYO-PRE and vinblastine was mediated through a mitochondrial pathway.

Summary: This study investigated the inhibitory effects of the natural product 2',4'-Dihydroxy-6'-methoxy-3',5'-dimethylchalcone (DMC) derived from Syzygium nervosum A. Cunn. ex DC. on various cancer cell lines, particularly cervical cancer. DMC displayed significant antiproliferative activity in different cervical cancer cell lines and induced DNA damage, cell cycle arrest, and apoptosis. The findings suggest that DMC has the potential to be developed as a therapeutic agent for antiproliferative cervical cancer treatment.

The preliminary screening suggests that the sooty molds have the potential to produce novel compounds and contribute to sustainable agriculture.

Summary: This study aimed to investigate the effects of perilla cake supplementation in a low-lysine diet on the meat quality of late-phase pigs. The results showed that adding perilla cake significantly increased intramuscular fat and marbling score, while reducing boiling loss and shear force value. However, it was found that perilla cake supplementation led to a significant increase in malondialdehyde levels in meat compared to the basal diet. Additionally, perilla cake supplementation significantly increased the level of alpha-linolenic acid in backfat and longissimus thoracis et lumborum muscle.

Summary: In this study, the bioactive compounds in shallot extract were evaluated for their hair growth-promoting activities. The results showed that phenolic compounds were the major constituents of the extract and exhibited anti-inflammatory effects. The extract also regulated the expression of genes related to hair growth pathways. These findings suggest that shallot extract has potential in promoting hair growth.

Summary: This study evaluates the efficiency of bovine sperm sexing using magnetic-activated cell sorting (MACS) by scFv antibody against Y-chromosome-bearing sperm (Y-scFv) coupled to magnetic microbeads and its effects on kinematic variables, sperm quality, and X/Y-sperm ratio. The results show that PY-microbeads are highly effective in separating Y-sperm from X-sperm with no negative impact on the quality of X-sperm. The technique produces high percentages of X-sperm and Y-sperm, making it a feasible method for bovine sperm sexing.

Summary: The aim of this study was to investigate the effects of KDML105 bran extract on gene expression involved in the hair cycle, as well as its antioxidant and anti-inflammatory activities. The extract contained compounds such as tocopherols, gamma-oryzanol, phytic acid, and phenolic compounds, which contributed to its antioxidant capacities. Treatment with the extract reduced nitric oxide production and downregulated SRD5A2 and TGFB1 expressions in HFDPCs, while upregulating CTNNB1 and VEGF genes. The findings suggest that KDML105 bran extract may play a role in promoting hair growth and angiogenesis, making it a promising source of anti-hair loss substances.

Summary: Alopecia and gray hair are common hair abnormalities with psychological impacts, and chemical treatments have distressing side effects. This study focused on the waste of rice cultivars from Thailand and evaluated their potential for hair revitalization. The BB4CMU rice bran oil (RBO) showed comparable melanin production induction to the standard drug theophylline, and it also exhibited inhibitory effects on steroid 5 alpha-reductase genes. Thus, BB4CMU-RBO could be a promising candidate for hair revitalization from natural resources.

Summary: Datura metel L. (thorn apple) was extracted using supercritical carbon dioxide technique and its biological activities were evaluated through various assays. The results suggest that supercritical carbon dioxide extraction could be a potential method for Datura metel extraction and wound closure.
