Summary: Primordial germ cells in the fetal testis transform into spermatogenic stem cells (SSCs) during the neonatal period, playing a crucial role in male fertility. The process of gonocyte to SSC transformation involves morphological and transcriptomic changes, as well as migration within the seminiferous tubules. Recent studies have highlighted the importance of mini-puberty, the temporal activation of the hypothalamic-pituitary-gonadal axis, in supporting gonocyte-to-SSC transformation. Understanding the significance of mini-puberty is essential for spermatogenesis research.

Summary: This systematic review analyzes the current evidence for assessing the efficacy of medical castration using biochemical markers. The available evidence suggests that the current method of measuring serum testosterone levels during medical castration is of low quality. Immunoassays were predominantly used instead of the recommended liquid chromatography with tandem mass spectrometry (LC-MSMS) method to measure low testosterone concentrations. Longitudinal studies measuring serum testosterone with LC-MSMS, serum-free testosterone, and luteinising hormone are needed to determine their clinical impact.

Summary: Recent studies have shown that 17 alpha-estradiol can extend the healthspan and lifespan of male mice through multiple mechanisms, making it a potential candidate for translation into humans. However, there is currently no established dosing paradigm for the treatment of aging and chronic disease in humans. This study aimed to assess the tolerability of 17 alpha-estradiol treatment and evaluate metabolic and endocrine responses in male rhesus macaque monkeys during a short treatment period. The results showed that the dosing regimens were well-tolerated and had mild benefits on metabolic parameters, but significant feminization effects were observed.

Summary: This study found that quercetin treatment can improve memory impairments caused by paradoxical sleep deprivation (PSD) in an animal model in a dose-dependent manner. Quercetin can restore the dynamic changes of the gamma band and significantly ameliorate the overactivation of microglia induced by PSD in the hippocampal CA1 region. Mechanistically, quercetin inhibits the TLR4/MyD88/NF-KB signaling pathway to suppress abnormal microglial activation.

Summary: This study compared the perioperative outcomes, overall survival, and treatment trends in patients with bladder cancer who underwent radical cystectomy and received either neoadjuvant or adjuvant chemotherapy. The findings suggest that neoadjuvant chemotherapy is associated with better overall survival and lower administration of granulocyte colony-stimulating factor when compared to adjuvant chemotherapy. Therefore, neoadjuvant chemotherapy should be considered as a treatment option for bladder cancer patients undergoing radical cystectomy.

The results showed that GBE increased the expression of steroidogenesis-related genes and proteins, and increased testosterone levels both in vitro and in vivo. Therefore, GBE might be a promising therapeutic or additive nutritional agent for improving men's health by increasing steroidogenesis or improving late-onset hypogonadism (LOH).

Summary: Multiple oral chemotherapeutic agents have been developed for use in combination with conventional androgen deprivation therapy for metastatic hormone-sensitive prostate cancer (mHSPC). A systematic review of randomized controlled trials found that many of these agents were more effective than the standard of care in terms of overall survival and progression-free survival. However, ADT + docetaxel with androgen receptor targeted agent (ARTA) triplet therapy was not the best treatment and requires further study.
