A Regulatory Variant in CCR6 Is Associated With Susceptibility to Antitopoisomerase-Positive Systemic Sclerosis

ObjectiveRecognition of the well-known pleiotropism of autoimmune genes supports the concept of a shared pathogenesis across autoimmune diseases such as rheumatoid arthritis (RA) and systemic sclerosis (SSc). Studies have reproducibly demonstrated an association between susceptibility to RA and polymorphisms of the CCR6 gene, a surface marker for Th17 cells, and the causal variant was recently identified. The present study was thus undertaken to investigate whether CCR6 polymorphisms could also be associated with susceptibility to SSc. MethodsTwelve tag single-nucleotide polymorphisms (SNPs) of CCR6, including the known RA-associated SNP rs3093023, were genotyped in a total of 2,411 SSc patients and 7,084 healthy individuals from 3 European populations (France, Italy, and Germany). Meta-analyses of the data were performed to assess whether an association exists between CCR6 polymorphisms and susceptibility to SSc or its main subtypes. Direct sequencing of DNA was performed to ascertain whether the functional dinucleotide polymorphism of CCR6 previously identified in RA (CCR6DNP) was also present in SSc. ResultsCombined analyses revealed an association between the rs10946216 SNP and SSc susceptibility (odds ratio [OR] 1.13, 95% confidence interval [95% CI] 1.05-1.21, adjusted P [P-adj] = 0.026). The rs3093023 A allele and rs10946216 T allele were in high linkage disequilibrium, and both were found to confer disease susceptibility in the antitopoisomerase-positive subset of SSc patients (OR 1.27, 95% CI 1.13-1.42, P-adj = 1.5 x 10(-3) and OR 1.32, 95% CI 1.17-1.48, P-adj = 9.0 x 10(-5), respectively, relative to healthy controls). Direct sequencing of the DNA of 78 individuals supported the hypothesis that the regulatory dinucleotide CCR6DNP could be the causal variant in SSc. ConclusionThe results of this study establish CCR6 as a new susceptibility factor for antitopoisomerase-positive SSc, as demonstrated in 3 European Caucasian populations, confirming the notion that SSc and RA could conceivably share autoimmune risk alleles. The results also suggest a potential role of the interleukin-17 pathway in SSc.

Summary: The study aimed to estimate the prevalence of long-term exposure to glucocorticoids (GCs) among SSc patients and identify factors associated with prescribing practices over time and across countries. The results showed that the utilization of GCs is widespread and long-term in SSc patients, with significant variations between countries and within-country. However, there has been a gradual decrease in their utilization.

Summary: This research aimed to identify candidate metabolite biomarkers of Metabolic Syndrome (MetS) and its associated risk factors. The study identified 56 specific metabolites related to MetS, with 13 of them being positively associated and 43 being negatively associated. The majority of these metabolites were associated with low HDL-C and hypertension. Additionally, one lipid metabolite, lysoPC a C18:2, was negatively associated with MetS and its five components. The study also revealed impaired metabolism of branched-chain and aromatic amino acids, as well as accelerated Glycine catabolism through metabolic networks.

Summary: This study used data from 2,325 population-based studies to analyze the height and BMI of children and adolescents aged 5-19 years in urban and rural areas from 1990 to 2020. The results showed that the height advantage of urban residents over rural residents has decreased in most countries, while it has amplified in sub-Saharan Africa. The BMI difference between urban and rural areas is small in most countries, with faster BMI growth in urban areas observed in some regions.

OTTERS: a powerful TWAS framework leveraging summary-level reference data

Summary: Most TWAS tools require individual-level eQTL reference data, making them not suitable for summary-level reference eQTL datasets. Hence, the development of TWAS methods that can utilize summary-level reference data is valuable. In this study, a TWAS framework called OTTERS is introduced, which adapts multiple polygenic risk score methods to estimate eQTL weights from summary-level data and performs omnibus TWAS. Both simulations and application studies demonstrate the practicality and power of OTTERS as a TWAS tool.

Associations of myosteatosis with disc degeneration: A 3T magnetic resonance imaging study in individuals with impaired glycaemia

Summary: This study evaluated the different associations of magnetic resonance imaging (MRI)-based paravertebral myosteatosis with lumbar disc degeneration in individuals with impaired glucose metabolism and normoglycaemic controls. The results showed a significant positive association between paravertebral myosteatosis and intervertebral disc degeneration in individuals with impaired glucose metabolism. No significant associations were found in the normoglycaemic controls. These findings emphasize the potential link between skeletal muscle and spinal-related diseases.

Summary: This study demonstrates the feasibility of deep learning-based image segmentation for the assessment of adipose tissue and spatial distribution of visceral and subcutaneous fat in a large population-based cohort in Germany. The results show significant differences in fat distribution between genders and age/body mass index-related displacements. Deep learning-based image segmentation provides a robust and fast quantification of adipose tissue and assessment of its spatial distribution in large cohort studies.

Associations between medium- and long-term exposure to air temperature and epigenetic age acceleration

Summary: Climate change poses a serious threat to human health worldwide, while aging populations increase. However, no study has ever investigated the effects of air temperature on epigenetic age acceleration. This study involved participants from two population-based studies in Germany and found that medium- and long-term exposures to high air temperature are associated with increases in epigenetic age acceleration. Overall, this study highlights the importance of considering climate change and temperature in relation to human health.
