Inhibition of Bone Morphogenic Protein 4 Restores Endothelial Function in db/db Diabetic Mice

Objective-Bone morphogenic protein 4 (BMP4) is involved in the development of endothelial dysfunction in hypertension. This study investigated whether the inhibition of BMP4 signaling improves endothelial function in db/db diabetic mice. Approach and Results-Male db/db mice were treated with noggin via osmotic pump infusion (1 mu g/[h.kg(-1)]) for 2 weeks. Adenovirus BMP4-short hairpin RNA was introduced via tail vein injection at a dosage of 10(9) pfu/mouse and its effects were examined 7 days after. Vasoreactivity was studied on wire and pressure myograph. Both noggin treatment and adenovirus BMP4-short hairpin RNA transduction improved endothelium-dependent relaxations in aortae and flow-mediated dilatation in mesenteric arteries of db/db mice. Ex vivo treatment with BMP4 inhibitors and adenovirus BMP4-short hairpin RNA rescued the impaired endothelium-dependent relaxations in db/db mouse aortae and reduced reactive oxygen species overproduction determined by dihydroethidium staining, CM-H(2)DCFDA fluorescence imaging, and chemiluminescence assay in db/db mouse aortae, and also in ex vivo cultured C57BL/6 mouse aortae or primary mouse aortic endothelial cells treated with high glucose. Likewise, activin receptor-like kinase 3 silencing by short hairpin RNA lentivirus improved endothelium-dependent relaxations in db/db mouse aortae accompanied by reactive oxygen species inhibition in endothelial cells. In addition, noggin reduced BMP4 upregulation in high-glucose-treated endothelial cells and in C57BL/6 mouse aortae and in aortae from db/db mice. Conclusions-Inhibition of BMP4/activin receptor-like kinase 3/reactive oxygen species signaling improved endothelial function in diabetic mice through limiting oxidative stress in endothelium. Inhibiting BMP4 cascade can become another potential therapeutic strategy against diabetic vascular dysfunction.

SOX4 is a novel phenotypic regulator of endothelial cells in atherosclerosis revealed by single-cell analysis

Summary: This study utilized single-cell RNA sequencing to explore the transcriptional landscape and heterogeneity of dysfunctional endothelial cells in atherosclerotic aortae. SOX4 was identified as a novel marker for atherosclerosis and was shown to promote atherogenesis and endothelial-to-mesenchymal transition. Hyperlipidemia-associated cytokines and oscillatory blood flow were found to upregulate SOX4 expression, while the anti-diabetic drug metformin suppressed SOX4 levels in endothelial cells.

This process promotes monocyte/macrophage adhesion to endothelium.

Risk of diabetes mellitus among users of immune checkpoint inhibitors: A population-based cohort study

Summary: This prospective cohort study analyzed cancer patients receiving immune checkpoint inhibitors in Hong Kong from 2013 to 2021, and found that the use of immune checkpoint inhibitors may lead to new-onset diabetes mellitus. Male patients were more likely to develop this risk, but there was no significant difference in risk between PD-1 inhibitors and PD-L1 inhibitors users.

Predictive value of neutrophil-to-lymphocyte ratio for atrial fibrillation and stroke in type 2 diabetes mellitus: The Hong Kong Diabetes Study

Summary: This study evaluated the predictive value of neutrophil-to-lymphocyte ratio (NLR) for ischemic stroke and atrial fibrillation (AF) in patients with type 2 diabetes mellitus. The results showed that NLR was significantly associated with an increased risk of new-onset ischemic stroke and AF. NLR had predictive value for ischemic stroke but lower predictive value for AF in Chinese type 2 diabetes patients.

Summary: Research found that cardiovascular diseases are the leading cause of death in type 2 diabetes, and current diabetic medications have limited effects on cardiovascular mortality. Experiments on mouse aortas showed that protocatechuic acid (PCA) can improve endothelial function and has antioxidant and anti-inflammatory effects. The protection on cardiovascular function is exerted through the Akt/eNOS pathway. Therefore, it is recommended for diabetic patients to consume an appropriate amount of PCA daily.

A self-indicating and antibacterial gelatine-chitosan blended hydrogel enabling real-time quality control and sustained bioactive agent delivery

Summary: In this study, a crosslinked blended hydrogel was generated using gelatine and chitosan with clusteroluminogenic properties, which not only exhibited intrinsic antibacterial properties and high tunability in delivery performance but also had a self-indicating capacity for quality control during hydrogel preparation. The release profiles of the agent-loaded gels followed the Higuchi model well, with the non-Fickian mechanism being the major mechanism of the release process. Our gels have high efficiency in agent loading and have great potential in bioactive agent delivery and related biomedical applications.

IL-4/STAT6 axis observed to reverse proliferative defect in SCA3 patient-derived neural progenitor cells

Summary: This study reveals that SCA3 patient-specific induced neural progenitor cells (iNPCs) exhibit proliferative defects and reduced autophagic expression compared to control. Furthermore, these SCA3 iNPCs undergo rapid senescence and do not survive subsequent passages. However, exposure to interleukin-4 (IL-4) can increase autophagic programs and reduce the proliferation defect in SCA3 iNPCs.

Effects of increasing light versus moderate-to-vigorous physical activity on cardiometabolic health in Chinese adults with obesity

The results showed that MVPA had more positive effects on body composition and cardiometabolic biomarkers compared to LPA.
