Human CD34+/KDR+ Cells Are Generated From Circulating CD34+ Cells After Immobilization on Activated Platelets

Objective-The presence of kinase-insert domain-containing receptor (KDR) on circulating CD34(+) cells is assumed to be indicative for the potential of these cells to support vascular maintenance and repair. However, in bone marrow and in granulocyte colony-stimulating factor (G-CSF)-mobilized peripheral blood, less than 0.5% of CD34(+) cells co-express KDR. Therefore, we studied whether CD34(+)/KDR+ cells are generated in the peripheral circulation. Methods and Results-Using an ex vivo flow model, we show that activated platelets enable CD34(+) cells to home to sites of vascular injury and that upon immobilization, KDR is translocated from an endosomal compartment to the cell-surface within 15 minutes. In patients with diabetes mellitus type 2, the percentage of circulating CD34(+) co-expressing KDR was significantly elevated compared to age-matched controls. When treated with aspirin, the patients showed a 49% reduction in the generation of CD34(+)/KDR+ cells, indicating that the level of circulating CD34(+)/KDR+ cells also relates to in vivo platelet activation. Conclusion-Circulating CD34(+)/KDR+ are not mobilized from bone marrow as a predestined endothelial progenitor cell population but are mostly generated from circulating multipotent CD34(+) cells at sites of vascular injury. Therefore, the number of circulating CD34(+)/KDR+ cells may serve as a marker for vascular injury. (Arterioscler Thromb Vasc Biol. 2011;31:408-415.)

Summary: This study aimed to investigate the association between changes in HDL composition and increased microvascular risk in individuals with type 2 diabetes mellitus. Differences in HDL composition were found between Dutch South Asian and Dutch white Caucasian individuals with diabetes, and these differences were associated with the incidence of microvascular complications. These HDL compositional differences could serve as ethnicity-specific biomarkers for type 2 diabetes.

Presence of immunogenic alternatively spliced insulin gene product in human pancreatic delta cells

Summary: Transcriptome analysis identified insulin-gene-derived transcripts in non-beta endocrine islet cells. Researchers studied alternative splicing of human INS mRNA in pancreatic islets and found an alternatively spliced INS product that is expressed in delta cells but not in beta cells. This variant encodes the complete insulin signal peptide and B chain, and its presence in delta cells may play a role in islet autoimmunity and pathology.

Summary: The aim of this study was to identify differentially expressed lncRNAs and mRNAs in whole blood of people with type 2 diabetes across five different clusters. The results showed that multiple lncRNAs and mRNAs were found to be differentially expressed among clusters, with particular significance in the MOD cluster. These findings contribute to our understanding of the molecular mechanisms underlying different clusters of type 2 diabetes.

Efficient Vascularization of Kidney Organoids through Intracelomic Transplantation in Chicken Embryos

Summary: Kidney organoids derived from human induced pluripotent stem cells resemble adult kidneys to a certain extent, but lack functional vasculature and limited maturation in vitro. Transplanting kidney organoids into the celomic cavity of chicken embryos induces vascularization and enhances maturation. This technique is efficient and enables transplantation and analysis of a large number of organoids.

Summary: Proliferative forms of glomerulonephritis are characterized by leukocyte influx, albuminuria, and loss of kidney function. The glycocalyx, comprised of heparan sulfate, plays a crucial role in glomerular inflammation. Exogenous glomerular glycocalyx and low-molecular-weight heparin reduce proteinuria and granulocyte/macrophage influx in experimental glomerulonephritis, as well as prohibit granulocyte adhesion to glomerular endothelial cells. The development of HS-based therapeutics for inflammatory glomerular diseases is justified by these findings.

Summary: Accumulating evidence suggests that endothelial dysfunction is involved in the progression of COVID-19. This study found that serum factors of severe COVID-19 patients may affect the glycocalyx and lead to endothelial dysfunction. The study also demonstrated that certain substances circulating in the plasma of COVID-19 patients in intensive care units are associated with endothelial glycocalyx degradation, barrier disruption, inflammation, and increased coagulation.

Summary: The prevalence of end-stage kidney disease (ESKD) is increasing in older patients, with cardiovascular disease (CVD) being the main cause of death. Circulating angiogenic miRNAs, angiopoietin-2 (Ang-2), and asymmetric dimethylarginine (ADMA) levels are associated with cardiovascular structure and function in ESKD patients. ADMA and miR-27a levels are higher in ESKD patients with coronary heart disease, while miR-27a levels are higher in patients with atrial fibrillation. These markers are strongly correlated with measures of cardiovascular structure and function.
