Increased levels of histones have been associated with COVID-19, increasing the risk of heart attack and stroke. Therefore, histones can be used as diagnostic biomarkers for COVID-19 and as predictive laboratory tools for targeted therapies.

Summary: The study investigated the role of extracellular histones on monocyte distribution width (MDW) and their effect on the release of cytokines by blood cells. The results showed that extracellular histones can induce changes in MDW and promote heterogeneity in monocytes. They also lead to the release of cytokines, especially G-CSF, IL-1 beta, IL-6, MIP-1 beta, and IL-8. These findings may have significant implications for predicting the risk of severe outcomes.

The influence of redox modulation on hypoxic endothelial cell metabolic and proteomic profiles through a small thiol-based compound tuning glutathione and thioredoxin systems

Summary: Reduction in oxygen levels is a key feature in the physiology of the bone marrow (BM) niche where hematopoiesis occurs. Antioxidant molecules can modulate the redox status of endothelial cells (ECs) in a hypoxic environment, promoting hematopoietic stem cell (HSC) maintenance. In this study, HUVEC cells exposed to low oxygen were treated with I-152, which increased glutathione levels and influenced the metabolic profiles interconnected with the glutathione system. Gene and protein expression analyses showed modulation of redox-related genes and upregulation of antioxidants. I-152 also reduced oxidative stress and downregulated certain cytokines in hypoxic ECs, potentially creating an in vitro BM niche conducive to functional HSC maintenance.

Platelet distribution width (PDW) as a significant correlate of COVID-19 infection severity and mortality

Summary: SARS-CoV-2 infection can cause a wide range of symptoms, and platelet indices have attracted great interest due to their involvement in COVID-19-related thromboinflammation. A systematic review found significantly elevated platelet distribution width (PDW) values in COVID-19 infected cohorts compared to controls, with higher PDW values observed in severe cases. These results suggest that platelets play a critical role in the immunothrombosis process of COVID-19, and PDW can serve as an indicator of platelet heterogeneity.

Summary: This study found that histones can significantly increase the monocyte distribution width (MDW) index in whole blood samples from healthy individuals when treated in vitro, similar to the changes found in patients with classic and viral sepsis. These changes are associated with histone-induced modifications of monocyte volume, cytoplasmic granularity, vacuolization, and nuclear structure. Therefore, histones may play a crucial role in the observed alterations of monocytes in acute classical and viral sepsis.
