Twenty-Five Novel Loci for Carotid Intima-Media Thickness: A Genome-Wide Association Study in >45 000 Individuals and Meta-Analysis of >100 000 Individuals

Ming Wai Yeung, Siqi Wang, Yordi J. van de Vegte, Oleg Borisov, Jessica van Setten, Harold Snieder, Niek Verweij, M. Abdullah Said, Pim van der Harst

Summary: Three genome-wide association studies were conducted in over 45,000 individuals, identifying 25 novel loci associated with cIMT. These findings provide insights into the genetic architecture underlying atherosclerosis and the shared biological mechanisms with cardiovascular diseases.

Large HDL particles negatively associate with leukocyte counts independent of cholesterol efflux capacity: A cross sectional study in the population-based LifeLines DEEP cohort

Clinical paper Routine reporting of grey-white matter differentiation in early brain computed tomography in comatose patients after cardiac arrest: A substudy of the COACT trial

K. O. Adriaansens, L. S. D. Jewbali, J. S. Lemkes, E. M. Spoormans, M. Meuwissen, M. J. Blans, P. van der Harst, B. J. W. Eikemans, G. B. Bleeker, A. Beishuizen, J. P. Henriques, A. van der Lugt, N. van Royen, C. A. den Uil

Summary: This study investigated the frequency and reporting methods of grey-white matter differentiation (GWR) in brain CT scan reports in comatose patients after out-of-hospital cardiac arrest (OHCA), and its association with prognosis. The study found that although GWR was frequently described in the reports, early abnormal GWR assessed qualitatively within 24 hours after return of circulation (ROSC) was a poor predictor of neurological prognosis.

Identification and single-base gene-editing functional validation of a cis-EPO variant as a genetic predictor for EPO-increasing therapies

Summary: By studying genetic variants and performing functional validation, we identified a variant in the EPO gene promoter region associated with EPO levels. Using genetic prediction and large-scale genome-wide association data, we found no significant evidence of increased risk of coronary artery disease, myocardial infarction, or stroke with genetically predicted long-term rises in EPO.

Summary: This study reveals that carriers of novel genetic variants of GPR146 have a favorable cardiometabolic risk profile. However, it remains unclear whether genetic or pharmaceutical inhibition of GPR146 can protect against atherosclerosis in humans.

Clonal haematopoiesis of indeterminate potential: associations with heart failure incidence, clinical parameters and biomarkers

Canxia Shi, Joseph Pierre Aboumsallem, Navin Suthahar, Aniek O. de Graaf, Joop H. Jansen, Isabelle A. van Zeventer, Valentina Bracun, Sanne de Wit, Elles M. Screever, Pieter F. van den Berg, Wouter C. Meijers, Ron T. Gansevoort, Stephan J. L. Bakker, Pim van der Harst, Herman H. W. Sillje, Gerwin Huls, Rudolf A. de Boer

Summary: This study aimed to analyze the association between clonal hematopoiesis of indeterminate potential (CHIP) and incident heart failure (HF) in a European population cohort. Results showed that CHIP mutations were correlated with age, smoking, hypertension, and cardiovascular biomarkers, but the frequency of CHIP was similar between individuals with incident HF and control participants. However, among people younger than 65 years, CHIP mutations were more frequently detected in the case cohort and significantly associated with new-onset HF.

Unified approach for solving exit problems for additive-increase and multiplicative-decrease processes

Remco van der Hofstad, Stella Kapodistria, Zbigniew Palmowski, Seva Shneer

Summary: This article analyzes a process that grows linearly in time and experiences downward jumps at Poisson epochs. Exit problems from fixed intervals and half-lines are considered, and explicit formulas for Laplace transforms are given using a unified first-step analysis approach.

Predictors of adverse diastolic remodeling in non-diabetic patients presenting with ST-elevation myocardial infarction

Summary: This study aimed to identify the factors leading to diastolic remodeling after STEMI. The results showed that female gender, multivessel coronary artery disease, and micro-vascular damage were important predictors of adverse diastolic remodeling. Additionally, older age and hypertension prior to STEMI may contribute to worse pre-existing diastolic function.

Electrocardiogram-based deep learning improves outcome prediction following cardiac resynchronization therapy

Summary: This study utilizes an explainable deep learning algorithm to predict the outcome of cardiac resynchronization therapy (CRT) using electrocardiogram (ECG) features. The algorithm outperforms current guideline ECG criteria and QRS(AREA) in predicting clinical endpoints. FactorECG, derived from a large dataset of ECGs, demonstrates superior discriminative ability without requiring additional clinical variables.

Entropy of labeled versus unlabeled networks

Summary: The labeling noise in networks may overpower the network structure. The common practice of using exchangeable models to reason about real-world networks with distinguishable nodes may introduce uncontrolled aberrations, suggesting a need for reexamination of the statistical foundations and key results of network science.

The role of inter-regional mobility in forecasting SARS-CoV-2 transmission

Martijn H. H. Schoot Uiterkamp, Martijn Goesgens, Hans Heesterbeek, Remco van der Hofstad, Nelly Litvak

Summary: This paper investigates the role of mobility in predicting the spread of SARS-CoV-2 and uses the case of The Netherlands as a study. By incorporating mobility into a transmission model, the study finds that considering inter-regional mobility generally improves the accuracy of forecasting, especially when municipalities have a relatively large amount of incoming mobility.
