Preparation of transgenic Dunaliella salina for immunization against white spot syndrome virus in crayfish

Although a white spot syndrome virus (WSSV) subunit vaccine could significantly enhance the immune response and benefit the shrimp host, its practical application is currently not feasible because of drawbacks in existing expression systems. We generated a transgenic Dunaliella salina (D. salina) strain by introducing the WSSV VP28 gene to produce a novel oral WSSV subunit vaccine. Following transformation of D. salina, VP28 gene expression was assessed by reverse transcription polymerase chain reaction (RT-PCR) assays, enzyme-linked immunosorbent assays (ELISAs), and western blot analysis. The RT-PCR results indicated that the VP28 gene was successfully expressed in D. salina cells. The presence of recombinant VP28 proteins with natural bioactivity was confirmed by western blot analysis and ELISA. Animal vaccination experiments indicated that transgenic D. salina can induce protection against WSSV by oral delivery in crayfish. Our findings indicate that the VP28 gene can be successfully expressed in transgenic D. salina and can be applied as an oral vaccine to protect crayfish against WSSV. We have demonstrated that it is feasible to produce an oral vaccine using D. salina, and thereby provide a new method for controlling other viral diseases in crustaceans.

Summary: In this study, a metal-free cascade dihalogenation-cyclization reaction was reported for the convenient preparation of dibrominated cis-tetrahydrobenzofuran-5(4H)-ones and cis-dihydrobenzopyran-6(5H)-ones. These products bear 1,4-dibromobut-1-enyl subunit in trans-trans and trans-cis form. The anticancer activity against MDA-MB-468 cells was also observed for the corresponding products.

Discovery of Novel Drug-like PHGDH Inhibitors to Disrupt Serine Biosynthesis for Cancer Therapy

Summary: PHGDH is an important enzyme in serine biosynthesis pathway, and its overexpression in malignant tumor cells makes it a potential target for cancer treatment. In this study, a series of novel PHGDH inhibitors were discovered using compound screening and structural optimization. The lead compound D8 showed strong enzymatic inhibitory activity, high binding affinity, and sensitivity to PHGDH gene amplification or overexpression. Further experiments revealed the binding site of D8 on PHGDH and its potential antitumor efficacy in a xenograft mouse model.

A characteristics analysis of carbon emission based on multi-dimensional carbon emission accounting methods and structural decomposition analysis: A case study of Beijing, China

Summary: This study established a multi-dimension long-term carbon emission analysis model to explore the characteristic of Beijing's embodied carbon emissions. The analysis revealed the contribution of socioeconomic factors in local and imported carbon emissions and identified the key contributors to carbon emissions. The study is expected to provide decision support for policymakers in formulating carbon mitigation policies and allocating carbon mitigation responsibilities from multiple perspectives.

Effects of platform's blockchain strategy on brand manufacturer's distribution strategy in the presence of counterfeits

Summary: This study establishes a theoretical model between a brand manufacturer and an e-commerce platform. The manufacturer aims to open a new online channel through the platform to attract more consumers, in addition to the traditional channel. However, the online channel faces the threat of counterfeits, and the platform can choose to adopt blockchain technology to address this issue. The main findings indicate that blockchain not only has an anti-counterfeiting effect but also has an incentive effect, where the platform can use blockchain to incentivize the manufacturer to open the online channel. Furthermore, both the supply chain and consumers can benefit from blockchain, and the impact of different channel characteristics on blockchain's value is analyzed.

Fixed-time prescribed performance optimization control for the speed and tension system of the cold strip rolling mill with output constraints

Summary: This article proposes a fixed-time prescribed performance optimization control method based on disturbance observers for the speed and tension system of the reversible cold strip rolling mill with output constraints, parameter perturbations, and load disturbance. The disturbance observers are constructed to estimate the system's unmatched uncertainties, and the observer errors can converge in fixed time. The designed controllers, combined with a time-varying logarithmic barrier Lyapunov function (TLBLF), command filtered backstepping approach, fixed-time control, and prescribed performance control, ensure the convergence of system states in fixed time and the constraints within predefined ranges.

Synergistic Effect of Energy Absorption and Adiabatic Temperature on the Microstructural Evolution and Mechanical Properties During High-Speed Impacts

Summary: High-speed impact at strain rates >10(3) s(-1) has common characteristics in energy absorption and adiabatic temperature rise. An ultra-high strength low alloy steel subjected to high-speed impact was studied, with a focus on energy absorption driven by thermo-kinetics phenomenon and its correlation with target plate thickness. Deformation characteristics were explained using an analytical model that relates absorbed energy and plate thickness. EBSD and FE analysis revealed the formation of adiabatic shear bands due to adiabatic temperature rise in specific zones, while the radial direction experienced increased stress resulting in a higher proportion of recrystallized grain structure. Micro hardness testing confirmed the differences in hardness between the distinct zones.

Coupled effects of sedimentary iron oxides and organic matter on geogenic phosphorus mobilization in alluvial-lacustrine aquifers

Summary: The biodegradation of organic matter and the dissolution of iron oxides play important roles in the release of geogenic phosphorus to groundwater. However, the combined effects of natural organic matter and iron oxides on phosphorus mobilization are not well understood. This study investigates sediment samples from boreholes in the Central Yangtze River Basin and finds that sediments with high phosphorus levels contain more bioavailable phosphorus, particularly iron oxide bound phosphorus and organic phosphorus, compared to sediments with low phosphorus levels. The study also reveals the formation of stable phosphorus-iron-organic matter complexes that inhibit the dissolution of iron oxides and the degradation of organic matter, thereby limiting phosphorus mobilization.

Mitochondrial genomic characterization of two endemic Chinese freshwater crabs of the genus Sinopotamon (Brachyura: Potamidae) and implications for biogeography analysis of Potamidae

Summary: In this study, the complete mitogenomes of Sinopotamon chishuiense and the nearly complete mitogenomes of S. wushanense were sequenced and analyzed. Novel patterns of gene rearrangement were detected, and the phylogenetic relationship was revealed. The results have significant implications for understanding the evolutionary history and taxonomic relationship of Sinopotamon.

Nature-inspired catalytic asymmetric rearrangement of cyclopropylcarbinyl cation

Summary: Researchers report an asymmetric rearrangement of cyclopropylcarbinyl cations using artificial organic catalysts, resulting in chiral homoallylic sulfide products with high yields and enantioselectivities (up to 99% enantiomeric excess). The reaction involves the dehydration of prochiral cyclopropylcarbinols catalyzed by a chiral SPINOL-derived N-triflyl phosphoramide catalyst, followed by trapping of the resulting cyclopropylcarbinyl cations by thione-containing nucleophiles. The synthetic utility of the process is demonstrated through subgram-scale experiments and downstream transformations.

Investigation of Electro-Thermal Performance for TreeFET from the Perspective of Structure Parameters

Summary: By using TCAD simulation, the electro-thermal properties of TreeFET are investigated and optimized by considering the interbridge width, nanosheet space, and nanosheet width. It is found that a smaller interbridge width mitigates short-channel effects and increases electron concentration, but also increases thermal resistance. A larger nanosheet space increases on-state current and compensates for gate drive loss, but results in longer conduction paths, limiting further improvements. Additionally, a suitable nanosheet width is required to lessen the decline of gate controllability induced by interbridge channels. Therefore, suitable geometry parameters should be selected to achieve a compromise between thermal and electrical performance.

Task Assignment of UAV Swarms Based on Deep Reinforcement Learning

Summary: UAV swarm applications are crucial for the future, but creating a dynamic and scalable assignment algorithm is a challenge. To address this, we propose the Ex-MADDPG algorithm, which improves robustness and scalability. Through simulations and real-world experiments, our results demonstrate the effectiveness and scalability of the Ex-MADDPG algorithm in handling various groups and tasks. This algorithm holds great promise for mission planning and decision-making in UAV swarm applications.

Optimization and understanding of corrosion inhibitors for cooling water system

Summary: This study aims to optimize the concentration and pH value of two corrosion inhibitors, nitrite-based and mixture of nitrite and molybdate, for carbon steel protection in closed cooling water systems. The study reveals that the mixture of nitrite and molybdate exhibits higher inhibition efficiency due to its double layer structure.

Preliminary study on diagnostic reference levels of whole-body PET-CT examinations in Jiangsu Province, China

Summary: This study surveyed whole-body positron emission tomography/computed tomography (PET-CT) examinations in Jiangsu Province and established diagnostic reference levels (DRLs). Nine hospitals in three regions of Jiangsu Province were included in the study. Various metrics were used to establish the DRLs, including activity duration product (ADP), activity per body weight (A(W)), dose-length product (DLP), and total effective dose (E-T). The DRLs for whole-body PET-CT examinations in Jiangsu Province were determined to be 922 mGy • cm (DLP), 4453 MBq.min (ADP), 5.22 MBq.kg(-1)(A(W)), and 19.8 mSv (E-T). The E-T for whole-body PET-CT examinations in Jiangsu Province was found to be higher than in other countries.

eDNA metabarcoding reveals differences in fish diversity and community structure in heterogeneous habitat areas shaped by cascade hydropower

Summary: This study used the efficient and noninvasive environmental DNA metabarcoding (eDNA) approach to survey the aquatic ecology of 12 river sections in the Wujiang River. The results revealed a decline in fish diversity and a change in fish community structure in the mainstream of the river, caused by anthropogenic factors such as hydropower and overfishing. The eDNA method was found to complement traditional methods and provide accurate information on fish composition.

Production of the antifungal biopesticide physcion through the combination of microbial fermentation and chemical post-treatment

Summary: In this study, we report on the successful preparation of 3R-torosachrysone (4), a tetrahydroanthracene precursor of physcion, in Aspergillus oryzae NSAR1 by heterologous expression of related genes mined from the phlegmacins-producing ascomycete Talaromyces sp. F08Z-0631. The conditions for converting 4 into physcion were studied and optimized, leading to the development of a concise approach for extracting high-purity physcion from the alkali-treated fermentation broth of the 4-producing A. oryzae strain.
