Objective: This cross-sectional study aimed to investigate the relationship among microbiological composition of dental plaque, sugar exposure and social factors, as well as the presence of visible plaque in preschoolers with different stages of early childhood caries. Design: A total of 169 children were clinically examined according to the World Health Organisation criteria + early caries lesions and were divided into three groups: caries-free (n = 53), early caries lesions-ECL (n = 56), and cavitated caries lesions-CCL (n = 60). The presence of clinically visible dental plaque on maxillary incisors was recorded. After that, dental plaque from all buccal and lingual smooth surfaces was collected and the number of colony-forming units of mutans streptococci and total microorganism, as well as the presence of lactobacilli was determined. Daily frequency of meals containing sugar was assessed by a diet chart whereas social factors were evaluated by a questionnaire. The data were analysed by chi-square test followed by multiple logistic regressions (alpha = 0.05, confidence interval = 95%). Results: High levels of mutans streptococci (OR = 2.28), high total sugar exposure (OR = 5.45) and presence of dental plaque (OR = 2.60) showed significant association with ECL (p <0.05). High total microorganism count (OR = 5.18), high solid sugar exposure (OR = 2.50) and the presence of lactobacilli (OR = 24.99) revealed significant association with CCL (p <0.05). Conclusions: These results support the conclusion that dietary sugar experience influenced the microbiological composition of dental plaque. Moreover, the early stage of caries is highly affected by mutans streptococci and visible dental plaque on maxillary incisors whereas cavities are strongly related to lactobacilli and total microorganism. Debonding of orthodontic appliance changes salivary physicochemical properties and favors regression of active caries lesions: A 13-week follow-up study

Andreia Alves Cardoso, Emerson Tavares de Sousa, Carolina Steiner-Oliveira, Marines Nobre-dos-Santos

Summary: This study investigated the effects of orthodontic appliance removal (OAR) combined with caries-preventive strategies and fluoride varnish treatments on salivary physicochemical properties, the activity of carbonic anhydrase VI (CA VIACT) and alpha-amylase (alpha-AML(ACT)), and the regression of active caries lesions (ACL). The results showed that this treatment significantly reduced the number of ACL, decreased the percentage of visible biofilm and sugar exposure, and increased salivary pH and alpha-AML(ACT) activity. However, salivary buffering capacity and CA VIACT activity remained stable throughout the follow-up.

Summary: This study describes a new protocol for polymicrobial oral conventionalization in mice. By inoculating specific pathogen free animals with wild mouse microbiota, the researchers were able to achieve colonization of a microbiota similar to that of wild animals for up to 80 days. This technique proves to be effective, low-cost, and easily reproducible.

Summary: Early childhood caries (ECC) is a common chronic disease in young children, characterized by atypical and fast progressive caries lesions. The severe form of ECC, known as S-ECC, can cause significant tooth damage and affect the child's quality of life. The development of ECC is driven by an imbalance between the oral microbiome and the host, mediated by high sugar consumption and poor oral hygiene. Understanding the microbiome in ECC is crucial for risk monitoring and the development of therapies to restore oral health.

Influence of salivary acquired pellicle on colour changes and minerals content of dentine after application of silver diamine fluoride followed by potassium iodide

I. B. Camatta, N. F. Peres, M. Z. D. Picolo, V. Cavalli, C. Steiner-Oliveira

Summary: This study evaluated the effect of salivary acquired pellicle (AP) on dentin darkening and caries arresting after using SDF + KI. The results showed that SDF and APSDF groups had no difference in color, while KISDF and APKISDF groups had significantly different colors compared to those without KI. SDF, APSDF, and APKISDF groups had less mineral loss and greater mineral recovery compared to C and KISDF groups. The presence of AP did not affect dentin color after SDF + KI, but played a protective role in modulating dentin mineral gain/loss. KI reduced dentin darkening and did not interfere with dentin de/remineralization process.
