Summary: This study investigated the effects of exosomes from human adipose-derived mesenchymal stem cells (adMSC-Exos) on extracellular matrix (ECM) remodeling in keloids. The results showed that adMSC-Exos inhibited the expression of collagen and fibronectin, disrupted collagen production and microvessel structure, and had an impact on the protein expression of certain factors involved in ECM production. These findings provide a new potential alternative for the treatment of keloids by targeting ECM production.

Summary: This study investigated the association of TGF-beta 1, 2, and 3 isoforms with plaque stability in atherosclerosis. The results showed that TGF-beta 2 is the most abundant isoform in human plaques and may maintain plaque stability by reducing inflammation and matrix degradation.

Summary: This study investigated the effects of osteomodulin (OMD) on the cell proliferation, migration, and extracellular matrix (ECM) synthesis of keloid-derived fibroblasts (KFs). The results showed that OMD was highly expressed in keloid tissues and was positively correlated with the activation of p38 MAPK signaling pathway. Silencing OMD inhibited the proliferation and migration of TGF-beta 1-treated KFs, as well as the expression of collagen and fibronectin; while overexpression of OMD had the opposite effects. These findings suggest that OMD may promote the hyperproliferation, migration, and excessive ECM synthesis in KFs through regulation of the p38 MAPK signaling pathway.

Summary: The study showed that corilagin has multiple effects on HSFs, including inhibition of collagen production, cell proliferation, cell migration, and suppression of HSF activation. Corilagin also significantly suppressed HS formation and collagen deposition in a rabbit ear scar model, as well as inhibited fibroblast proliferation and alpha-SMA expression in vivo. Additionally, western blot analysis revealed that corilagin downregulated the protein levels of TGF-beta 1 and TGF-beta receptor type I (TGF beta RI), thus affecting the levels of p-smad2/3, MMPs, and TIMP1.

Summary: The article discusses the mechanisms and functional interactions regulating BMP bioavailability in cellular microenvironments, emphasizing how structural disturbances in ECM proteins may lead to dysregulated BMP signaling as an underlying cause for connective tissue disease pathways. Additionally, current mechanistic concepts of how aberrant BMP signaling drives connective tissue destruction in inherited and chronic diseases are provided.

Summary: In this study, it was found that UCHL1 was significantly upregulated in keloid fibroblasts and played a role in the pathological process of keloid by promoting collagen I and alpha-SMA expression. UCHL1 expression was also found to be promoted by M2 macrophages via TGF-beta 1. These findings expand our understanding of the pathogenesis of keloid and provide potential therapeutic targets for the disease.
