Summary: This study investigates the embryonic toxicogenomic responses induced by Zinc pyrithione (ZPT) in zebrafish. The results show that ZPT treatment causes embryogenesis toxicity, including irregular cell division and rearrangement, delayed differentiations of eyes and notochords, and defects in epiboly and germ ring formation and somite segmentation. ZPT exposure also alters gene expression during early embryonic development, leading to morphological abnormalities and metabolic dysfunctions. Additionally, ZPT induces oxidative stress, apoptosis, endoplasmic reticulum (ER) stress, and autophagy.

Summary: This study aimed to compare different techniques for measuring sperm nuclear DNA fragmentation (sDF) and examine their relationship with boar reproductive value, classical spermiogram parameters, and reproductive results of the doses in sows. The findings indicate a significant correlation between sperm chromatin stability assay (SCSA) and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay results. However, there was no correlation between sDF and fertility direct boar effect (DBE) index, total or progressive sperm motility. TUNEL measure showed a positive correlation with abnormal acrosomes and sperm chromatin dispersion test (SCD, Halomax (R)) measure showed a positive correlation with total sperm morphological abnormalities. No significant correlations were found between fertility or prolificacy results and sDF results with the different techniques.

Summary: This study analyzed eight techniques assessing chromatin damage in pig sperm and found that the extent of chromatin damage is significantly correlated with the percentage of sperm with fragmented DNA in direct methods, but not in indirect ones. The results suggest that there are differences in interpreting sperm chromatin damage between indirect and direct techniques.

Summary: This study investigated the adverse effects of Zinc Pyrithione (ZPT) on mouse oocyte maturation and quality. The results showed that ZPT compromised oocyte maturation by disrupting meiotic cell cycle and causing cytoskeletal defects. In addition, ZPT altered epigenetic modifications and resulted in zinc accumulation, which further impaired the quality of oocytes.

Summary: The European Union has recently banned the use of zinc pyrithione (ZPT) in cosmetic products due to its potential health risk, which has raised the need for a reliable and convenient detection method. This study presents a highly sensitive method using surface-enhanced Raman spectroscopy (SERS) for on-site one-step detection of ZPT in real-life samples. The SERS sensor, produced by electrochemically treating commercial silver needles, allows for easy sampling and rapid identification of ZPT in shampoos within one minute using a portable Raman spectrometer.

Summary: Zinc pyrithione (ZPT) is widely used as an antifouling, antibacterial, and antifungal agent, but it has been shown to be toxic to marine life. However, there is limited research on the toxicity and toxicological mechanism of ZPT in the reproductive systems of aquatic organisms. Our study found that ZPT exposure caused various abnormalities in zebrafish sperm and testicular tissue, including decreased testosterone levels, sperm deformities, decreased sperm concentration and motility, and DNA damage in testicular tissue. We also identified potential mechanisms of ZPT-induced testicular injury through RNA-seq analysis of differentially expressed genes (DEGs).

Various factors can affect the results, and challenges and clinical implications of TUNEL in male infertility evaluation are briefly discussed.
