Pharmaceutically-active compounds are regularly and widely released into the aquatic environment in an unaltered form or as metabolites. So far, little is known about their potential detrimental effects on algae populations which can ultimately impact nutrient cycling and oxygen balance. For our analysis, the common microalga Pseudokirchneriella subcapitata (P. subcapitata) was exposed to a mixture of 13 drugs found in Italian wastewaters and rivers. Traces of pharmaceuticals investigated were detected in treated algal cells, except for cyclophosphamide and ranitidine, indicating that these algae are able to absorb pharmaceutical pollutants from the environment. The effects of the treatment were investigated by Amplified Fragment Length Polymorphism (AFLP) assessment of DNA damage and 2-DE proteomic analysis. While no genotoxic effect was detected, proteomic analysis showed that algae are sensitive to the presence of drugs and that, in particular, the chloroplast is affected. (C) 2010 Elsevier B.V. Perspectives and challenges associated with the determination of new psychoactive substances in urine and wastewater-A tutorial

L. Bijlsma, R. Bade, F. Been, A. Celma, S. Castiglioni

Summary: New psychoactive substances (NPS) are emerging as legal substitutes to conventional illicit drugs, causing public health concerns. Monitoring NPS use is challenging, but determining NPS in pooled urine and wastewater shows great potential. However, this method also presents analytical challenges and limitations.

Proteomic analysis reveals how pairing of a Mycorrhizal fungus with plant growth-promoting bacteria modulates growth and defense in wheat

Summary: Plants rely on their microbiota for improving nutrition and stress tolerance, with some plant growth-promoting bacteria and arbuscular mycorrhizal fungi showing potential benefits for wheat growth. Plant responses to different microbial combinations can lead to specific phenotypic and proteomic changes, influencing defense mechanisms and immune responses. The presence of arbuscular mycorrhizal fungi seems to be crucial for promoting plant growth and development in synthetic microbial communities.

Symbiotic responses of Lotus japonicus to two isogenic lines of a mycorrhizal fungus differing in the presence/absence of an endobacterium

Francesco Venice, Matteo Chialva, Guido Domingo, Mara Novero, Andrea Carpentieri, Alessandra Salvioli di Fossalunga, Stefano Ghignone, Angela Amoresano, Candida Vannini, Luisa Lanfranco, Paola Bonfante

Summary: By analyzing Lotus japonicus roots colonized by the fungal line with bacteria (B+) and by the cured line (B-) through deep dual-mRNA sequencing and proteomics, it was found that L. japonicus activates its symbiotic pathways regardless of the presence of endobacteria, but perceives the two fungal lines as different physiological entities. Significant differences were observed in respiratory pathways and lipid biosynthesis between the mycorrhizal plants colonized by the B+ and B- lines, indicating a reduced capacity in antioxidant metabolism and symbiotic lipid biosynthesis in the cured line.

Summary: This study focused on 28 NPS stimulants and detected 11 of them, with eutylone and 3-methylmethcathinone (3MMC) being the most commonly found and with the highest mass loads, indicating international popularity. Older generation stimulants like para-methoxyamphetamine (PMA), methylone, and mephedrone were also detected, showing the dynamic nature of the NPS market. Areas in New Zealand were found to have the highest loads of NPS stimulant consumption, demonstrating the potential public health risks associated with these substances.

Efficient extraction of carrageenans from Chondrus crispus for the green synthesis of gold nanoparticles and formulation of printable hydrogels

Milena Alvarez-Vinas, Noelia Gonzalez-Ballesteros, M. Dolores Torres, Lucia Lopez-Hortas, Candida Vanini, Guido Domingo, M. Carmen Rodriguez-Arguelles, Herminia Dominguez

Summary: This study investigates the sustainable extraction and depolymerization of carrageenan from seaweed using hydrothermal treatment with subcritical water. The results show that operating at 140 degrees C yields the highest extraction rate, and the crude carrageenan can be effectively obtained through methods such as ultrafiltration and ethanol precipitation. Additionally, it is found that the crude carrageen fraction can be used to prepare gel materials with excellent mechanical properties.

Proteomics as a tool to decipher plant responses in arbuscular mycorrhizal interactions: a meta-analysis

Guido Domingo, Candida Vannini, Marcella Bracale, Paola Bonfante

Summary: The symbiotic relationship between plants and arbuscular mycorrhizal (AM) fungi leads to significant changes in plant metabolism, which are regulated by various molecular mechanisms. This study conducted a comprehensive analysis of AM-modulated proteins at both the local (roots) and systemic (shoots/leaves) levels using proteomics. The analysis identified a total of 4262 differentially abundant proteins and revealed a systemic reprogramming of translation, with a central role played by ribosomal translational apparatus.
