The oxytetracycline (OTC) is an antibiotic widely used in the aquaculture, principally in the treatment of infections caused by the pathogenic Gram-negative bacteria. In the present study, the consequences of the oral administration of OTC on the main humoral (natural haemolytic complement activity, IgM levels and seric peroxidase level) and cellular (respiratory burst, intracellular peroxidase level and phagocytosis) immune parameters of gilthead seabream (Sparus aurora. L) were evaluated after 7, 14 and 21 days of treatment Furthermore, the expression levels of immune-associated genes (MHC-II alpha., C3, IL-1 beta, COX-2 and beta-defensin) were analysed by real-time polymerase chain reaction in head-kidney and gut. Three experimental groups were established: control group (fed with non-supplemented OTC food) and experimental groups (4 and 8 mg OTC g(-1) feed, respectively) (N = 30). The results reveal a decrease of the natural haemolytic complement activity to the higher dose of OTC administered. By contrast, after 7 days of treatment, fish fed the lowest concentration of OTC were observed to have a statistically significant increase in the percentage of phagocytic cells, while the phagocytic capacity and respiratory burst increased after 14 days of treatment for both concentrations of OTC. No significant effects were observed after 21 days of treatment for both concentrations of OTC with regard to the values obtained in the control group. In head-kidney and gut, the dietary administration of OTC not caused statistically significant variation, only in C3 gene expression at 7 days in gut. The present results suggest that after dietary administration of OTC, the humoral and cellular innate immune parameters of gilthead seabream are depressed and stimulated respectively, and induce up- or down-regulation of the immune-relevant genes analysed depending on the concentration of OTC supplied in diet. Implication of mucus-secreting cells, acidophilic granulocytes and monocytes/macrophages in the resolution of skin inflammation caused by subcutaneous injection of λ/κ-carrageenin to gilthead seabream (Sparus aurata) specimens

Jose Carlos Campos-Sanchez, Enrica Vitarelli, Francisco A. Guardiola, Diana Ceballos-Francisco, Jose Maria Garcia Beltran, Antonio Ieni, Maria Angeles Esteban

Summary: This study evaluated local inflammation mechanisms in the skin of gilthead seabream induced by carrageenin, finding an increase in skin mucus-secreting cells and acidophilic granulocytes after injection. Gene expression of various markers and proinflammatory genes was also up-regulated post-injection. Results indicate the chronological involvement of important immune cells in resolving inflammation in the fish skin.

Summary: In this study, Schiff base derived organosilatranes with pyridine moiety were successfully synthesized using a simple method. The structure of the compounds was determined through various spectroscopic studies. The chemosensor exhibited high selectivity and sensitivity towards Zn2+ ions, as evidenced by significant changes in UV-vis and fluorometric spectra. Immobilizing the organosilatrane on silica nanoparticles surface enhanced its binding capacity for Zn2+ ions and showed potential for water treatment applications. Additionally, the synthesized organosilatranes showed promising pharmacokinetic profiles and cytotoxicity in biological field applications.

Implication of adipocytes from subcutaneous adipose tissue and fatty acids in skin inflammation caused by λ-carrageenin in gilthead seabream (Sparus aurata)

Jose Carlos Campos-Sanchez, Daniel Gonzalez-Silvera, Xu Gong, Richard Broughton, Francisco A. Guardiola, Monica B. Betancor, Maria Angeles Esteban

Summary: The role of adipocytes and fatty acids in carrageenan-induced skin inflammation was studied in gilthead seabream. Results showed that injection of lambda-carrageenin led to an increase in adipocyte area at 3 hours post-injection and an increase in adipocyte frequency at 6 hours post-injection. Analysis of lipid classes revealed increased levels of free fatty acids and sphingomyelin in the injected fish compared to the control group. The fatty acid profile of the fish skin also showed changes, with an increase in saturated fatty acids and n-6 polyunsaturated fatty acids, and a decrease in monounsaturated fatty acids, n-3 polyunsaturated fatty acids, and minor fatty acids at 6 hours after injection. Evaluation of eicosanoids indicated an increase in hydroxyeicosatetraenoic acid in the injected fish compared to the control group. These findings demonstrate the contribution of adipocytes and fatty acids to the inflammatory response in gilthead seabream skin.
