Alnus peptides modify membrane porosity and induce the release of nitrogen-rich metabolites from nitrogen-fixing Frankia

Actinorhizal plant growth in pioneer ecosystems depends on the symbiosis with the nitrogen-fixing actinobacterium Frankia cells that are housed in special root organs called nodules. Nitrogen fixation occurs in differentiated Frankia cells known as vesicles. Vesicles lack a pathway for assimilating ammonia beyond the glutamine stage and are supposed to transfer reduced nitrogen to the plant host cells. However, a mechanism for the transfer of nitrogen-fixation products to the plant cells remains elusive. Here, new elements for this metabolic exchange are described. We show that Alnus glutinosa nodules express defensin-like peptides, and one of these, Ag5, was found to target Frankia vesicles. In vitro and in vivo analyses showed that Ag5 induces drastic physiological changes in Frankia, including an increased permeability of vesicle membranes. A significant release of nitrogen-containing metabolites, mainly glutamine and glutamate, was found in N-2-fixing cultures treated with Ag5. This work demonstrates that the Ag5 peptide is central for Frankia physiology in nodules and uncovers a novel cellular function for this large and widespread defensin peptide family.

Biotechnological potential of Kocuria rhizophila PT10 isolated from roots of Panicum turgidum

Structure of surface polysaccharides affects symbiotic nitrogen fixation in Frankia alni ACN14a

Exploring the Phytobeneficial and Biocontrol Capacities of Endophytic Bacteria Isolated from Hybrid Vanilla Pods

Summary: In this study, endophytic bacterial strains were isolated from pods of hybrid vanilla plants from Madagascar and genetically characterized. Results showed that bacteria belonging to the genus Bacillus and Curtobacterium exhibited beneficial capabilities such as phosphate solubilisation, free nitrogen fixation, and phytohormone and siderophore production. Some strains also showed potential for biocontrol of vanilla pathogenic fungi. Spectral imaging revealed the production of lipopeptides by bacterial strains confronted with the pathogenic fungi.

Direct measurement of the aerotactic response in a bacterial suspension

DNA Methylation in Ensifer Species during Free-Living Growth and during Nitrogen-Fixing Symbiosis with Medicago spp

Summary: DNA methylation plays important roles in immunity and cellular processes regulation in Ensifer bacteria, especially in nitrogen-fixing symbiosis with legumes. Six genome-wide methylated motifs were identified using single-molecule real-time sequencing, with five of them being strain-specific. The GANTC motif recognized by the cell cycle-regulated CcrM methyltransferase was methylated in all strains. The methylation of GANTC motifs increased progressively from ori to ter regions in actively dividing cell cultures, while near full genome-wide GANTC methylation was observed in the early stage of symbiotic differentiation, followed by a moderate decrease in overall methylation and a progressive decrease in chromosomal GANTC methylation in later stages of differentiation. These findings suggest dysregulated and constitutive CcrM activity during terminal differentiation may be a driving factor for endoreduplication of terminally differentiated bacteroids.
