Summary: The new immuno-PCR method utilizes TotalSeq antibodies containing DNA conjugated oligos for high sensitivity and specificity profiling of multiple EV surface proteins simultaneously. This method has been successfully applied to cell and serum samples, and a micro-size exclusion chromatography method has been developed to detect EV surface proteins in as little as 10 μl of human serum when combined with immuno-PCR. Overall, this approach allows for rapid detection of multiple EV markers from small sample volumes in a single tube.

Summary: This study aimed to develop a multiplex reverse transcriptase polymerase chain reaction (m-RT-PCR) method for simultaneous detection of important avian viral pathogens. Specific primers were designed and synthesized using conserved segments of viral genetic sequences, and recombinant vectors were synthesized as positive controls. The d-optimal approach was used to optimize PCR conditions. The m-PCR assay showed high specificity, sensitivity, and reproducibility in detecting and evaluating viral infection in birds, with the ability to detect 8 target genes and identify 8 viral pathogens from infected samples. The test also demonstrated that live animal oral swabs were as effective as tissue sampling from dead animals, making it a valuable tool for viral infection analysis in poultry.

Summary: High rates of new cervical cancer cases and deaths are related to the limitation of regular cervical cancer screening in low- and middle-income countries. A new multiplex recombinase polymerase amplification assay was developed to detect 20 high-risk and 14 low-risk types of HPV, as well as specific genes related to cervical cancer development. The assay showed promising results in terms of specificity, diagnostic accuracy, speed, cost, and safety.

Summary: A multiplex-nested PCR (M-nested PCR) targeting mpt64 (Rv1980c) + IS6110 was designed for the detection of Mycobacterium tuberculosis (Mtb) DNA in urine, endometrial biopsies, and menstrual blood samples of UGTB patients. The M-nested PCR showed a lower detection limit and higher sensitivities compared to conventional M-PCR. Incorporation of an internal amplification control helped confirm true PCR-negative results. GeneXpert MTB/RIF assay had lower sensitivity but high specificity compared to M-nested PCR. The M-nested PCR shows promise as a potential diagnostic tool for UGTB.
