Developing new immunotherapy strategies involving therapeutic targeting of HIF-1 molecules associated with hypoxia may enhance the clinical effectiveness of immunotherapy. Targeting hypoxia presents a potential opportunity to improve the clinical benefit of cancer immunotherapy.

Summary: This study aimed to analyze and evaluate the dynamic changes in HIF-1 alpha concentration in serum exosomes during bacterial peritonitis. A new colorimetric method to quantitate these changes was established. The results showed that the serum exosomal HIF-1 alpha levels increased in the early stage of bacterial peritonitis, reached a peak at 24-48 hours, and then gradually decreased. Intervention treatment reduced the abdominal infection extent, HIF-1 alpha concentration in serum exosomes, and the serum pro-inflammatory factors levels. These findings suggest that serum exosomal HIF-1 alpha can be used as a biomarker for predicting and monitoring the pathological process of bacterial peritonitis.

Summary: This study demonstrates that canagliflozin can suppress prostate cancer cell proliferation and enhance their sensitivity to radiotherapy. It achieves this by blocking mitochondrial respiration, promoting AMPK activity, inhibiting MAPK and mTOR-p70S6k/4EBP1 pathways, activating cell cycle checkpoints, inhibiting proliferation partly through HIF-1α suppression, and mediating transcriptional reprogramming of metabolic and survival pathways. Canagliflozin may have potential in combination with radiotherapy for the prevention and treatment of prostate cancer.

Summary: In this study, a novel PHD2 gene, SpPHD2, was cloned and identified in the mud crab Scylla paramamosain. The mRNA expression of SpPHD2 was widely distributed across all examined tissues, and the protein was mainly localized in the cytoplasm. Silencing SpPHD2 in vivo upregulated SpHIF-1 alpha and downstream genes, while overexpression of SpPHD2 in vitro dose-dependently reduced SpHIF-1 alpha transcriptional activity, indicating the crucial role of SpPHD2 in regulating SpHIF-1 alpha.

Summary: Hypoxia, or inadequate oxygen supply, is a characteristic of the tumor microenvironment and is associated with a poor prognosis and resistance to therapy. HIF-alpha, specifically the HIF-1 alpha and HIF-2 alpha subunits, play a crucial role in mediating the transcription of important proteins involved in tumor progression. While HIF-1 alpha has been extensively studied, the role of HIF-2 alpha is often overlooked. HIF-2 alpha can enhance the aggressive behavior of tumors, promoting metastasis, tissue remodeling, angiogenesis, and the upregulation of cancer stem cell factors. This review highlights the structure, function, hypoxic response, and roles of HIF-2 alpha in cancer progression, as well as discussing current therapeutic strategies and future directions.

Functional studies are needed to confirm the role of these genetic variants in the development of phenotypic abnormalities in individuals with Ollier disease and Maffucci Syndrome.

Summary: This study established a safe and effective method to downregulate the expression of HIF-1 alpha in hepatic stellate cells (HSCs) and treat liver fibrosis. By delivering HIF-1 alpha small interfering RNA (HIF-1 alpha-siRNA) via VA-PEG-modified CNs-based nanosheets encapsulated with GQDs, the expression of HIF-1 alpha can be decreased. Additionally, the nanosheets can trigger water splitting under near-infrared (NIR) irradiation, improving the hypoxic liver fibrosis site and achieving better therapeutic effects.
